摘要
目的:探讨美洲大蠊及其不同提取物对酒精致小鼠肝损伤的保护作用。方法:将小鼠随机分为正常对照组、模型组、多烯磷脂酰胆碱组(135 mg/kg)、原粉100、200 mg/kg剂量组、95%醇提100、200 mg/kg剂量组、75%醇提100、200 mg/kg剂量组、水提100、200 mg/kg剂量组。采用白酒灌胃的方式分别建立小鼠急性酒精性肝损伤和小鼠慢性酒精性肝损伤模型。测定各组小鼠ALT、AST含量水平,以及肝组织超氧化物歧化酶(SOD)活性,丙二醇(MDA)含量,观察小鼠肝脏病理变化。结果:给药组均能显著降低酒精致小鼠肝损伤血清中ALT、AST活性和肝脏组织中MDA的活性;显著升高肝脏组织中的SOD的活性;减轻小鼠肝脏病理损伤。水提组和原粉组小鼠血清中ALT、AST活性和肝组织中MDA、SOD活性与不同浓度醇提组比较有显著性差异,水提组与原粉组小鼠血清中ALT、AST活性和肝组织中MDA、SOD活性比较无差异。结论:美洲大蠊及其提取物对酒精性肝损伤具有一定的保护作用,但存在药效差异,其中原粉组和水提物的效果优于醇提两组,且同组间保护效果存在剂量依赖性。
        Objective: To explore the protective effect of Periplaneta americana and its different extracts on liver function induced by alcohol-induced liver injury in mice. Methods: The mice were randomly divided into normal control group,model group,positive drug group,insect powder suspension low and high(100,200 mg/kg) dose groups,95% alcohol extract low and high(100,200 mg/kg) dose groups,75% alcohol extract low and high(100,200 mg/kg) dose groups,water extract low and high(100,200 mg/kg) dose groups. The model of acute alcoholic liver injury and chronic alcoholic liver injury in mice were established by means of liquor gavage. The levels of ALT and AST,the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in liver tissue were measured. The pathological changes of liver were observed under light microscope. Results: The activity of ALT and AST(P < 0. 01,P < 0. 05) in serum and the activity of MDA(P < 0. 01,P < 0. 05) in liver tissue of mice induced by alcohol,and to ncreases SOD(P < 0. 01,P < 0. 05) activity in liver tissue reduce liver liver damage. The activity of ALT and AST and the activity of MDA and SOD in the liver tissue of the mice were significantly different from those of the different groups(P< 0. 01,P < 0. 05). There was no significant difference in serum ALT and AST activity and MDA activity between liver tissue and water suspension group(P > 0. 05). Conclusion: Periplaneta americana and its extracts have protective effect on alcoholic liver injury,among which the effect of powder group and water extract is the best. This study provides a basis for the subsequent treatment of alcoholic liver injury and its dosage form.
