摘要
目的:探讨回生胶囊对二甲基苯蒽(DMBA)诱导的骨髓增生异常综合征(myelodysplastic syndrome,MDS)大鼠模型的影响。方法:将60只SD大鼠随机分为正常组、模型组、环孢素组、回生胶囊高剂量组、低剂量组,每组12只。除正常组外,其他大鼠均采用化学诱变剂二甲基苯蒽建立MDS模型,于实验第14天各组开始给药,连续30 d。观察各组大鼠一般情况,病态造血,全自动血细胞分析仪检测外周血细胞,流式细胞仪检测外周血中CD3~+、CD4~+、CD8~+、CD4~+/CD8~+水平,ELISA法检测外周血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)水平。结果:模型组外周血和骨髓均见不同程度的病态造血,外周血中白细胞、红细胞、血红蛋白、血小板、CD3~+、CD4~+、CD4~+/CD8~+水平显著降低(P<0.01),TNF-α、IL-6水平显著升高(P<0.01);与模型组相比,各给药组均可不同程度地改善病态造血,外周血象、T淋巴细胞水平显著升高(P<0.01或P<0.05),TNF-α、IL-6水平显著降低(P<0.01或P<0.05)。结论:回生胶囊可通过改善MDS模型大鼠病态造血,升高外周血象、调节外周血T淋巴细胞水平,减少TNF-α、IL-6的释放,发挥抗肿瘤作用,且药物剂量与疗效呈正相关。
Objective: To investigate the effects of Huisheng capsule on rats with myelodysplastic syndrome(MDS)induced by dimethyl benzanthracene(DMBA).Methods:Sixty rats were randomly divided into normal group,model group,cyclosporine group,high-dose group of Huisheng capsule and low-dose group of Huisheng capsule,with 12 rats in each group.Except for the normal group,other rats were establish the MDS model by the chemical mutagen DMBA,the rats of each group were treated with different drugs on the 14 th day and treated continuously for 30 days.The general condition of rats in each group was observed.The pathological hematopoiesis was observed by microscopes. The peripheral blood was detected by automatic blood cell analyzer. The levels of CD3~+,CD4~+,CD8~+ and CD4~+/CD8~+ in peripheral blood were detected by flow cytometry. TNF-α and IL-6 were detected by ELISA.Results:Compared with the normal group,the model group showed three types of pathological hematopoiesis. The levels of white blood cells,red blood cells,hemoglobin,platelets,CD3~+,CD4~+,CD4~+/CD8~+ in peripheral blood were significantly decreased(P<0.01),TNF-α and IL-6 levels significantly increased(P <0.01). Compared with the model group,each treatment group could improve pathological hematopoiesis in different degrees,significantly increase peripheral blood and T lymphocyte levels(P <0.01 or P <0.05),and significantly reduce TNF-α and IL-6 levels(P <0.01 or P <0.05). Conclusion:Huisheng capsule could exert anti-tumor effect by improving the bone marrow pathological hematopoiesis,increase the peripheral blood picture,regulate the ratio of peripheral blood T lymphocytes,reduce the release of TNF-α and IL-6. And the drug dose was positively correlated with the efficacy.
