摘要
目的 探讨非小细胞肺癌(NSCLC)肿瘤及癌旁组织CD4~+和CD8~+T细胞上程序性死亡分子-1(PD-1)的表达及意义。方法 采用流式细胞术检测79例NSCLC患者肿瘤组织及78例NSCLC患者癌旁组织中CD4~+和CD8~+T细胞上PD-1的表达百分比,并分析其与NSCLC患者临床病理特征相关性。结果 肿瘤组织PD-1~+细胞占所有细胞百分比显著高于癌旁组织,差异有统计学意义(P<0.01)。肿瘤组织CD4~+和CD8~+T细胞表达水平显著高于癌旁组织,差异有统计学意义(P<0.01)。肿瘤组织CD4~+和CD8~+T细胞上PD-1表达水平显著高于癌旁组织,差异有统计学意义(P<0.01)。肿瘤>3 cm患者CD4~+和CD8~+T细胞上PD-1表达水平高于肿瘤≤3 cm患者,差异有统计学意义(P<0.05)。肺鳞癌患者CD8~+T细胞上PD-1表达水平高于肺腺癌患者,差异有统计学意义(P<0.05),而肺鳞癌患者CD4~+T细胞上PD-1表达水平与肺腺癌患者间差异无统计学意义(P>0.05)。PD-1表达在不同性别、年龄、病理分期、有无淋巴结转移间差异无统计学意义(P>0.05)。结论 NSCLC患者肿瘤组织CD4~+和CD8~+T细胞上PD-1表达水平明显高于癌旁组织,并与肿瘤大小具有相关性,可能为肺癌的研究提供参考。
        Objective To explore the expression and significance of programmed death-1(PD-1) on CD4~+ and CD8~+T cells in tumor tissues and adjacent tissues of patients with non-small cell lung cancer(NSCLC). Methods The expressions of PD-1 on CD4~+ and CD8~+T cells in the tumor tissues of 79 NSCLC patients and the adjacent tissues of 78 NSCLC patients were detected by flow cytometry analysis, and the correlations between PD-1 expressions and clinicopathological characteristics of NSCLC patients were analyzed.Results Compared with adjacent tissues,the expression level of PD-1~+ cells in tumor tissues was significantly higher statistically(P<0.01).The expression level of CD4~+ and CD8~+T cells in tumor tissues was significantly higher statistically(P<0.01).The expression level of PD-1 on CD4~+ and CD8~+T cells in tumor tissues was significantly higher statistically(P<0.01).The expressions of PD-1 on CD4~+ and CD8~+T cells in patients with tumor diameter >3 cm were higher than those with tumor diameter≤3 cm,the difference was statistically significant(P<0.05).The expressions of PD-1 on CD8~+T cells in patients with lung squamous cell carcinoma were higher than those with lung adenocarcinoma, and the difference was statistically significant(P<0.05), while the expressions of PD-1 on CD4~+T cells were not statistically significant(P>0.05).The expressions of PD-1 were not related to the gender, age, pathologic stages and lymph node metastasis(P>0.05).Conclusion The expression level of PD-1 on CD4~+ and CD8~+T cells in tumor tissues is significantly higher than that in adjacent tissues and correlated with tumor size, which may provide a reference for research of lung cancer.
