摘要
目的:阐述白介素17受体(IL-17R)mRNA在恒河猴不同组织中的分布与丰度。方法:建立实时荧光定量PCR方法,检测免疫、消化和生殖等系统的各组织中IL-17RA mRNA的水平,并检测了肠道IL-17RC mRNA的水平及恒河猴、非洲绿猴和食蟹猴PBMC中IL-17RA和IL-17RC mRNA的表达水平。结果:IL-17RA mRNA在所有检测过的组织中均有表达,但不同器官系统的表达水平存在显著差异(P<0.05);IL-17RC mRNA在大肠中的表达水平高于小肠;恒河猴PBMC中IL-17RA mRNA水平显著高于非洲绿猴(5倍)和食蟹猴(3倍),但PBMC中IL-17RC mRNA水平在3种动物中均低于检测下限。结论:IL-17RA广泛分布于恒河猴各种组织中,表达水平存在明显的组织差异和种属差异。
        Objective: To delineate the distribution and abundance of interleukin 17 receptor mRNA in normal tissues of rhesus macaques. Methods: Tissue samples were collected from different systems including lymphoid system,digestive system and genital system and the mRNA levels of IL-17 RA and IL-17 RC were examined by real time RT-PCR. Meanwhile the levels of IL-17 RA and IL-17 RC mRNA in the PBMCs of rhesus macaques,African green monkeys and cynomolgus monkeys were examined. Results: IL-17 RA mRNA was expressed in all the tested tissues and the levels were significantly different among these systems( P<0. 05). IL-17 RC mRNA level in the large intestine was higher than that in the small intestine. IL-17 RA mRNA level in rhesus macaque PBMCs was 3 and 5 times higher than that of cynomolgus monkeys and African green monkeys respectively,while IL-17 RC mRNA in the PBMCs was below the limit of detection. Conclusion: IL-17 RA is widely distributed in rhesus macaques; the expression level is different among different tissues and species.
