摘要
为了缩短优良赤松的品种选育时间,对赤松组培苗瓶内抗病性方法进行了研究。结果表明:以0.2%硫酸链霉素连续表面消毒松材线虫2次,茎尖注射接种400条/株线虫至半木质化赤松无菌苗,培养基表面添加0.1%多菌灵10mL,接种后进行DCR固体培养是较为理想的人工接种方法,可为今后进行松树瓶内抗病性测定提供参考。
For reducing breading time on excellent variety selection of Pinus densiflora,the research method on disease resistance on tissue cultured P.densiflora in jar was studied.The results showed that the ideal method of artificial inoculation was:twice surface disinfection with 0.2% streptomycin sulphate on Bursaphelenchus xylophilus,vaccination injection with concentration of 400 B.xylophilus per plant on stem tip of P.densiflora sterile seedlings with half lignification,10 mL carbendazim with concentration of 0.1% on the medium surface,solid DCR culture after inoculation.This method could provide relevant references for measurement of disease resistance on tissue cultured pine tree seedlings in jar.
