摘要
目的:探讨格列美脲对链脲佐菌素(STZ)诱导阿尔茨海默病(AD)模型小鼠认知行为的影响。方法:将30只昆明小鼠随机分为空白对照组、模型组及格列美脲3组,每组10只,模型组与格列美脲组小鼠侧脑室注射STZ,建立小鼠阿尔茨海默病(AD)模型,格列美脲组持续给药30 d,采用水迷宫实验,检测格列美脲对STZ-ICV模型小鼠认知行为的影响。结果:Morris水迷宫:定位航行试验中,AD模型组平均逃避潜伏期和路径长度较对照组明显增加,格列美脲治疗组平均逃避潜伏期和路径长度较AD模型组明显减少(P <0. 05);空间探索试验中,AD模型组穿越隐匿平台次数较对照组明显减少,格列美脲治疗组穿越隐匿平台次数较AD模型组显著增加(P<0. 05)。结论:格列美脲对STZ诱导的AD模型小鼠的学习记忆能力有明显的改善作用。
        Objective: To investigate the effect of glimepiride on the cognitive behavior of AD mice induced by streptozotocin(STZ). Methods: 30 Kunming mice were randomly divided into a blank control group,a model group,and three groups(10 mice in each group). STZ was injected into the lateral ventricle of mice in the model group and the glimepiride group to establish a mouse AD model. The glimepiride group was continuously administered for 30 days,and the effect of glimepiride on the cognitive behavior of STZ-ICV model mice was tested by water maze experiment. Results:Morris water maze: in the locational navigation test,the average escape latency and path length in the AD model group were significantly increased compared with the control group,and the average escape latency and path length in the glimepiride treatment group were significantly reduced compared with the AD model group(P < 0. 05). In the space exploration experiment,The Times of crossing the hidden platform in the AD model group were significantly reduced compared with the control group,and The Times of crossing the hidden platform in the glimepiride treatment group were significantly increased compared with the AD model group(P < 0. 05). Conclusions: Glimepiride significantly improved the learning and memory ability of stz-induced AD model mice.
