摘要
目的探讨生理性深层海水(PDSW)对45.0℃高温环境动物耐受性的影响及机制。方法将取自我国海南省海域的深层海水进行制备,制成PDSW,检测所含有的部分元素。20只昆明小鼠,雌雄各半,分为对照组和实验组,对照组小鼠饮用自来水,实验组饮用PDSW,连续15d,然后把各组小鼠放入45.0℃高温环境下饲养,记录每只小鼠死亡时间,直至各组小鼠全部死亡为止。并取每只小鼠脑、肺、心、肝和肾做病理检查。结果无论是雌性小鼠还是雄性小鼠,实验组的小鼠在45.0℃高温下的生存时间比对照组的小鼠长,差异有统计学意义(P<0.05)。并且雌性小鼠的生存时间要比雄性小鼠的生存时间长,差异有统计学意义(P<0.05)。组织学分析表明PDSW对实验小鼠的脑、心、肺、肝、肾等器官均有保护作用。Western blot检测发现,实验组小鼠肝组织HSP72表达较对照组强,差异有统计学意义(P<0.05)。结论 PDSW可以增强小鼠对45.0℃高温环境的耐受性,其机制考虑与PDSW促进热休克蛋白表达有关。
Objective To explore the effects of physiological deep-sea water(PDSW)on hyperthermal tolerance of Kunming(KM)mice in the 45.0 ℃ environment.Methods Deep-sea water from the south Chinese sea was processed,and the metallic elements dissolved in the DSW were analysed.The mice were randomly divided into 2groups:the control group received tap water;the experimental group treated with PDSW for 15 d.And then the mice were fed in the 45.0℃conditions.The survival time and histomorphometric analyses of the brain,lung,heart,liver and kidney were investigated.Results The survival time in PDSW-fed group was significantly longer than that of the control group(P<0.05).Moreover,histomorphometric analyses showed that PDSW could protect the brain,lung,heart,liver and kidney of KM mice from the 45.0℃conditions.The results of western blot revealed that expression of HSP72 of liver tissues for PDSW-fed group substantially increased,when compared with the control mice(P<0.05).Conclusion PDSW could improve hyperthermal tolerance of KM mice,which maybe in the relation with expression of HSP72 promoted by PDSW.
