摘要
目的探讨帕瑞昔布在大鼠肺缺血再灌注(LIR)损伤的作用及对肺内水通道蛋白-1(AQP-1)的影响。方法24只雄性成年SD大鼠随机分为3组:假手术组(C组)、缺血再灌注组(I组)和帕瑞昔布组(P组)。建立单侧LIR模型,检测肺湿/干重比值(W/D),观察肺组织病理变化,检测肺组织内TNF-α、IL-6含量,AQP-1表达情况。结果 LIR后,肺组织内TNF-α、IL-6明显增多(P<0. 05),肺湿/干重比值也增加(P<0. 05);与I组相比,以上指标在P组中均降低(P<0. 05)。肺组织形态学检测显示,LIR造成肺组织损伤严重,但P组较I组明显减轻。与C组相比,缺血再灌注明显抑制肺组织内AQP-1表达(P<0. 05);但P组表达水平较I组高(P<0. 05)。结论帕瑞昔布可抑制肺缺血再灌注引起的炎症反应,增加AQP-1表达,从而减轻损伤。
        Objective To investigate the effects of parecoxib on lung ischemic reperfusion( LIR) injury and aquaporin-1( AQP-1) in rats. Methods Twenty-four male adult SD rats were randomly divided into three groups: sham group( group C),ischemic reperfusion group( group I) and parecoxib group( group P). The unilateral LIR model was established to detect the lung wet/dry weight ratio( W/D),observe the pathological changes of lung tissues,and detect the expression of TNF-,IL-6 and AQP-1 in lung tissues. Results After LIR,TNF-a and IL-6 in lung tissue increased significantly( P<0. 05),and the lung wet/dry weight ratio increased( P<0. 05). Compared with group I,the above indexes were lower in group P( P<0. 05). The morphological examination of lung tissue showed that LIR caused severe lung injury,but the injury in group P was significantly less than that in group I.Compared with group C,ischemic reperfusion significantly inhibited the expression of AQP-1 in lung tissue( P <0. 05),but the expression level in group P was higher than that in group I( P<0. 05). Conclusion Parecoxib can inhibit the inflammatory response induced by lung ischemic reperfusion and increase the expression of AQP-1,thus reducing the injury.
