摘要
目的:建立以芦丁为对照品的留兰香(Herba Mentha spicata)总黄酮含量测定方法。方法:通过紫外扫描比较了芦丁对照品和留兰香提取物在亚硝酸钠-硝酸铝-氢氧化钠和三氯化铝2种显色体系下的吸收特性,确定一种检测方法作为留兰香总黄酮的测定较优方法。结果:亚硝酸钠-硝酸铝-氢氧化钠显色法更适合留兰香总黄酮含量的测定。标准曲线回归方程y=13.291x-0.011 5(R=0.999 8),芦丁的浓度和吸光度在8.008~48.050μg/mL范围内线性关系良好,平均加样回收率为97.4%,RSD为2.5%,重现性、稳定性和精密度均较好。结论:亚硝酸钠-硝酸铝-氢氧化钠显色法简便、快速、准确,可作为留兰香中总黄酮含量测定方法。
        Objective:This study was designed to develop a quantitative determination method of the total flavonoids from Herba Mentha spicata using rutin as a standard substance for further develpoment and utilization. Methods:Confirming which was the best detection method as determination of total flavonoid from Herba Mentha spicata,the rutin and Herba Mentha spicata extract(total flavonoids)was comparative analyzed by UV scanning method with NaNO_2-A l(NO_3)_3-NaOH chromogenic system and AlCl_3 chromogenic system,respectively. Results:The method of NaNO_2-Al(NO_3)_3-NaOH chromogenic system was accurate,with good precision,stability and repeatability for the determination of total flavonoids from Herba Mentha spicata. It was confirmed that the rutin had a linear relationship with absorbance in the range of 8.008 μg/ml~48.05 μg/ml(y=13.291 x-0.0115,R=0.999 8).The average recovery was 99.71%(RSD =1.32%). The average recovery was 97.4% with RSD of 2.5%. Conclusion:This method of NaNO_2-Al(NO_3)_3-NaOH chromogenic system was easy to operate,fast and accurate,and it is suitable to detect the total flavonoids from Mentha spicata L.
