游离棉酚降解产物四甲氧基棉酚致小鼠精子畸形效应的生殖毒性研究

摘要
目的研究游离棉酚降解产物四甲氧基棉酚的生殖毒性。方法采用小鼠精子畸变实验,将60只小鼠随机分为5组,空白组(40mg/kg,超纯水)、阳性对照组(40mg/kg,环磷酰胺)、四甲氧基棉酚的高(180mg/kg)、中(90mg/kg)和低(45mg/kg)剂量组,每组小鼠灌胃体积(20mL/kg体质量),每天1次,连续给药5d。取小鼠副睾制成精子涂片观察和计算各组小鼠精子畸形率。结果与正常小鼠精子相比,四甲氧基棉酚可导致小鼠精子形成香蕉型、胖头型和双尾型畸形类型。与空白组相比,阳性对照组小鼠精子畸形率为(8.20±0.31)%,四甲氧基棉酚高剂量组小鼠精子畸形率为(4.40±0.51)%、中剂量组小鼠精子畸形率为(3.00±0.33)%、低剂量组小鼠精子畸形率为(1.80±0.44)%,差异有统计学意义(P<0.05)。结论游离棉酚降解产物四甲氧基棉酚可导致小鼠精子畸形,具有生殖毒性。
        Objective To investigate the reproductive toxicity of the free gossypol degradation product tetramethoxy gossypol.Methods Sperm aberration was tested in mice.60 mice were randomly divided into 5 groups:blank group(40 mg/kg,ultrapure water),positive control group(40 mg/kg,cyclophosphamide),and tetramethoxy cotton in the high(180 mg/kg),middle(90 mg/kg),and low(45 mg/kg)dose groups,the volume of each group of mice(20 mL/kg.BW)was administered once a day,continuously drug for 5 days.The mice secondary testis was taken to make sperm smears observation and the sperm deformity rate was calculated by SPSS 17.0.Results Compared to normal sperm,tetramethoxy gossypol results in the formation of spermatozoa in to styles of banana,fat head and twin tails.Compared with the blank group,the rate of sperm abnormality in the positive control group was(8.2±0.31)%,the sperm deformity rate of the high dose group of tetramethoxy gossypol was(4.4±0.51)%,the sperm abnormality rate in mice was(3.0±0.33)%in the medium-dose group and(1.8±0.44)%in the low-dose group and the difference was statistically significant(P <0.05).Conclusion Tetramethoxyl gossypol as the degradation products of free gossypol can cause sperm deformity in mice,and its toxicity increases with the increase of concentration,and it has reproductive toxicity.
