葛根芩连汤对溃疡性结肠炎小鼠模型肠黏膜内NF-κB、TNF-α、MFG-E8及occludin表达的影响

摘要
目的探讨葛根芩连汤对溃疡性结肠炎(UC)小鼠模型肠黏膜内核因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、乳脂肪球表皮生长因子8(MFG-E8)及紧密连接蛋白(occludin)表达的影响。方法将40只balb/c小鼠分为A组(空白对照组,蒸馏水,n=10)、B组[模型组,葡聚糖硫酸钠(DSS)+蒸馏水,n=10]、C组[柳氮磺吡啶(SSZ)组,DSS+SSZ,n=10]及D组(葛根芩连汤组,DSS+葛根芩连汤,n=10)。观察并记录相应药物灌胃7d时的疾病活动指数(DAI)评分和组织损伤评分,测量、计算结肠长度、结肠质量及脾指数,检测UC小鼠模型肠黏膜内NF-κB、TNF-α、MFG-E8、occludin蛋白表达情况。结果B、C、D组建模第7d时,DAI评分、建模第8d时组织损伤评分均大于A组,且B组大于C、D组(P <0. 05),C、D组上述评分比较,差异无统计学意义(P> 0. 05)。建模第8d时,A、C、D组结肠长度大于B组,结肠质量、脾指数小于B组(P <0. 05),且A、C、D上述指标检测结果比较,差异无统计学意义(P> 0. 05);四组结肠黏膜NF-кB、TNF-α平均灰度值水平及MFG-E8、occludin蛋白相对表达量均显示A>C>D>B,MFG-E8、occludin平均灰度值水平及NF-кB、TNF-α蛋白相对表达量均显示A Objective To investigate the effects of Gegen-Qinlian decoction on the expression of nuclear factor-κB( NF-κB),tumor necrosis factor-α( TNF-α),milk fat globule epidermal growth factor 8( MFG-E8) and occludin in intestinal mucosa of mouse model of ulcerative colitis( UC).Methods Forty balb/c mice were randomly divided into group A( blank control group,distilled water,n = 10),group B( model group,dextran sodium sulfate( DSS) and distilled water,n = 10),group C( salazosulfapyridine( SSZ)group,DSS+SSZ,n = 10) and group D( Gegen-Qinlian decoction group,DSS and Gegen-Qinlian decoction,n = 10).The disease activity index( DAI) score and tissue injury score on the 7 thday of gavage administration of corresponding drugs were observed and recorded.The colon length,colon mass and spleen index were measured and calculated.The expressions of NF-κB,TNF-α,MFG-E8 and occludin protein in intestinal mucosa were detected.Results The DAI score on the 7 thday of modeling and the tissue injury score on the 8 th day of modeling in the groups B,C and D were all higher than those in the group A,and the scores in the group B were higher than those in the groups C and D( P < 0. 05),but there was no significant difference in the scores between the groups C and group D( P >0. 05).On the 8 thday of modeling,the colon length in the groups A,C and D was greater and the colon mass and spleen index were smaller than those in the group B( P < 0. 05),but there was no significant difference in the above indexes among the groups A,C and D( P > 0. 05).On the 8 thday of modeling,the average levels of gray value of NF-κB and TNF-α and the expression levels of MFG-E8 and occludin in colon mucosa among the four groups showed A>C>D>B,and the average gray value levels of MFG-E8 and occludin and the expression levels of NF-кB and TNF-α all showed that A
