摘要
研究3-羟基丁酸对HeLa癌细胞生长与凋亡的影响.采用四唑盐(MTT)比色法测定细胞活力;AO/EB荧光染色观察细胞凋亡与坏死的形态学变化;单细胞凝胶电泳检测细胞DNA的损伤;流式细胞仪检测细胞凋亡;蛋白质免疫印迹法(Western Blot)检测凋亡蛋白的表达.结果表明:3-羟基丁酸能够抑制HeLa癌细胞增殖;使HeLa癌细胞DNA受到损伤;通过caspase依赖性细胞凋亡途径诱导HeLa癌细胞凋亡;在HeLa癌细胞与人脐带正常细胞共培养条件下,3-羟基丁酸诱导HeLa癌细胞先于人脐带正常细胞凋亡.实验为肿瘤的生酮饮食疗法和酮体疗法提供了理论依据.
        To study the effects of 3-hydroxybutyrate on growth and apoptosis of HeLa cells, cell viability was detected by MTT. The morphological alterations of apoptosis or necrosis in HeLa cells were stained with AO/EB. HeLa cell DNA damage was analyzed by single cell gel electrophoresis(SCGE) test and the cell apoptosis was analyzed by flow cytometry. The expression of apoptotic protein was determined by Western Blot. The results showed that 3-hydroxybutyrate could inhibit the proliferation and damage the DNA of HeLa cells. The 3-hydroxybutyrate induced HeLa cancer cell apoptosis through caspase-dependent cell apoptosis pathway. In the co-culture condition, the 3-hydroxybutyrate induced HeLa cells to apoptosis before human umbilical cord cells. The results would provide a theoretical basis for the treatment of tumors with ketogenic diet and 3-hydroxybutyrate.
