摘要
目的应用Vevo2100高分辨率小动物超声评价异丙肾上腺素(ISO)诱导小鼠心肌缺血模型心功能实时变化与心脏病理学的关系,以及与心电图相比对模型成功与否的判断价值。方法 30只C57BL/6雄性小鼠随机分为两组,分别连续5d皮下注射生理盐水和ISO,于第7天、第14天、第21天行超声心动图检查、心电图检查及相关病理学染色。结果与NS组比较,ISO组病理学染色观察到心肌胶原容积分数明显升高;超声心动图检查发现,ISO组小鼠心脏射血功能明显降低且差异有统计学意义。心脏左室射血分数与心肌胶原容积分数和炎症评分均呈明显负相关。ISO开始注射后第7天,心电图监测证实均出现明显心肌缺血改变,同时进行超声心动图检查,经ROC曲线分析显示,超声检测左室射血分数(LVEF)预测第21天心肌纤维化的灵敏度和特异度均增高,且特异度明显高于心电图监测结果。结论高分辨率小动物超声能够准确评价ISO诱导的小鼠心肌缺血模型心功能和结构的动态变化,并在评价模型成功与否方面优于心电图监测。
        Objective Vevo 2100 small animal echocardiography was used to evaluate the relationship between cardiac pathological changes and real-time cardiac function of isoproterenol-induced myocardial ischemia mouse model,and the diagnostic value whether the model success or not compare with Electrocardiogram.Methods Thirty C57BL/6mice were randomly divided into two groups which subcutaneous inject ISO and normal saline respectively.Echocardiography,Electrocardiogram and pathological staining were performed at days 7,14 and 21follwing injection respectively.Results Pathological staining observed the collagen volume fraction of ISO group were significantly increased compare with control group.Electrocardiogram illustrates obvious ischemic changes.The echocardiography demonstrates the heart function of ISO group mouse were significantly decreased compare with control group,the difference is statistically significant.Correlation analysis of Echocardiography and pathological staining reveal ejection fraction was significant negative correlated with collagen volume fraction and inflammation score.ROC curve analysis showed that the sensitivity and specificity of LVEF predict day 21 myocardial fibrosis examed by echocardiography was increased compare with electrocardiogram confirmed myocardial ischemia.Conclusions The high-resolution small animal echocardiography could evaluate dynamic changes of cardiac function and structure in ISO-induced myocardial ischemia model in mice,and is better than electrocardiogram in predicting the mouse model successfully or not.
