生附子煮散工艺及与传统饮片煎出效果对比研究

摘要
目的优选生附片煮散工艺,并与附子传统饮片煎煮液中毒性成分和有效成分进行比较,探索其"减毒存效"作用机理,为附子临床应用提供指导原则。方法采用HPLC法测定附子煎煮液中6种单、双酯生物碱的质量分数,并以质量分数和干膏率为评价指标,考察生附片煮散主要影响因素(粒度、浸润时间、煎煮时间、煎煮次数),优选出生附片最佳煮散工艺;比较生附片煮散饮片和附子传统饮片(生附片和黑顺片)煎煮液中成分溶出差异。结果生附片最佳煮散工艺为:5~10目生附片煮散饮片加入400 m L清水,浸泡20 min,武火煮沸,文火煎煮30 min,滤过,即得。生附片中双酯型生物碱质量分数较高,需要经过60 min煎煮后才消失完全;生附片煮散饮片仅需煎煮30 min,其双酯型生物碱就基本消失,且其单酯型生物碱和干膏率分别是生附片煎煮液的1.63倍和1.26倍,黑顺片的4.22倍和3.68倍。结论生附子煮散工艺具有毒性低,成分溶出高的特点,且操作简单、稳定可控,是一种较为理想煎煮工艺。
        Objective To optimize the process of boiling powder of Shengfupian,to compare the difference of content of toxic components and active ingredients in the decoction with boiling powder and traditional herbal pieces,and explore its mechanism of " attenuation and effectiveness", which will provide guiding principles for clinical application of Aconite Lateralis Radix. Methods HPLC was used to determine the content of six kinds of alkaloids of single,and the content of the alkaloids and yield of dry extract were used as evaluation indexes to investigate the the main influencing factors of aconite decoction( particle size,infiltration time, decocting time, Decoction times), and the best boiling process was optimized. The difference of content of boiling powder and traditional herbal pieces was compared. Results The best process of boiling powder was as follows: the particle size was 5-10 meshes,and 400 m L fresh water was added,soaked for 20 min,steamed with high heat,decocted for 30 min,filtered and obtained. The content of diester alkaloids in Shengfupian was higher, and the diester-type alkaloids completely disappeared after60 min boiling,and the diester-type alkaloids in the decoction pieces disappeared only after decocting for30 min. The diester-type alkaloids almost disappeared, Compared with decoction of Shengfupian. The content of mono-alkaline alkaloids and yield of dry extract were 1. 63 and 1. 26 times, compared with decoction of Heishunpian,the content of mono-alkaline alkaloids and yield of dry extract were 4.22 and 3.68 times. Conclusion There was low toxicity and high dissolution of components about process of boiling powder,which was an ideal boiling process because of its simplicity,stability and controllability.
