摘要
目的:小花清风藤茎叶自然混合不同溶剂提取物对小鼠急性酒精性肝损伤的保护作用研究,并探讨其可能的机制。方法:实验小鼠随机分为9组,即正常组、模型组、水飞蓟素组(200mg/kg)、茎叶自然混合水提取物低、高剂量组、茎叶自然混合50%乙醇提取物低、高剂量组、茎叶自然混合95%乙醇提取物低、高剂量组。给药组分别灌胃给予相应剂量的药物,正常组和模型组分别灌胃给予等体积的0.5%羧甲基纤维素钠,1次/d,连续2周。给药1周后,于每次灌胃给药后4h,模型组和给药组灌胃给予12mL/kg的白酒(56%),正常组小鼠灌胃给予同体积蒸馏水,2次/d(上午8点,下午4点各1次),连续1周。实验期间自由饮水和进食,每天观察实验动物行为及体征。末次给药的次日,取材进行实验。取肝称重,计算肝质量指数;取血进行总胆固醇(TC)、甘油三酯(TG)含量检测;取肝组织匀浆检测谷丙转氨酶(ALT)、谷草转氨酶(AST)和谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量,另取肝组织进行组织形态学分析。结果:小花清风藤茎叶自然混合不同溶剂提取物可降低血液中TC、TG含量和肝组织中ALT、AST活性及MDA含量,升高肝组织中SOD和GSH-Px活性,从而减轻酒精对肝组织的氧化损伤,改善肝脏的组织病理学形态;不同溶剂提取物的作用效果由高到低依次为茎叶自然混合水提组、茎叶自然混合50%乙醇提组、茎叶自然混合95%乙醇提组,且呈现明显的量效关系。结论:小花清风藤茎叶自然混合的不同溶剂提取物对急性酒精性肝损伤小鼠具有显著的保护作用,其作用机制可能与其抗氧化作用有关。
        Objective:To observe the protective effect of different solvent extracts of natural mixture of stems and leaves in Sabia.parviflora Wall.ex Roxb on acute alcoholic liver injury in mice,and explore its possible mechanism.Methods:The experimental mice were randomly divided into 9 groups,namely,normal group,model group,silymarin group(200 mg/kg),water extract low and high dose groups of natural mixture of stems and leaves in Sabia.parviflora Wall.ex Roxb,50% ethanol extract low and high dose groups of natural mixture of stems and leaves in Sabia.parviflora Wall.ex Roxb,and 95% ethanol extract low and high dose groups of natural mixture of stems and leaves in Sabia.parviflora Wall.ex Roxb.The administration groups were given the corresponding dose drugs,the normal and model groups were given the equal volume of 0.5% sodium carboxymethyl cellulose,once a day for 2 weeks.After 1 weeks of continuous administration,the model group and the administration group were given 12 mL/kg alcohol(56%),the normal group mice were given the same volume of distilled water,twice a day(8 AM and 4 PM)for 1 week.Mice were freely access to water and food during the experiment.The behaviors and signs of experimental animals were observed everyday.At the end of experiment,the weights and mass indexs of liver were calculated,the levels of total cholesterol(TC)and triglyceride(TG)in blood and activity of alanine transarninase(ALT),glutamic-oxalacetic transaminase(AST),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and content of malondialdehyde(MDA)in the liver tissue were measured,and liver histopathological observations were analyzed.Results:The different solvent extracts of the natural mixed stems and leaves in Sabia.parviflora Wall.ex Roxb could decrease the content of TC and TG in the serum,activity of ALT,AST and the content of MDA in liver tissue,increase the levels of SOD and GSH-Px in liver tissue,thus reduce the oxidative damage of liver tissue and improve the liver histopathology,restore liver function.The effect of different solvent extracts from high to low was the water extract,50% ethanol extract and 95% ethanol extract of natural mixture of stems and leaves,and showed a significant quantity effect relationship.The aforementioned results confirmed its mechanism was related to its antioxidant activity.Conclusion:The different solvent extracts of the natural mixed stems and leaves in Sabia.parviflora Wall.ex Roxb have a significant protective effect on acute alcoholic liver injury in mice,and its mechanism may be related to its antioxidant effect.
