柴胡龙骨牡蛎汤对心肌梗死合并抑郁大鼠药效学作用的实验研究

摘要
目的:观察柴胡龙骨牡蛎汤对心肌梗死后伴发抑郁大鼠的药效学作用。方法:将68只大鼠随机分为正常组、模型对照组、西药组和中药组。给予模型对照组、西药组和中药组大鼠先后制造心肌梗死和抑郁症模型,造模成功后,正常组、模型对照组大鼠灌服生理盐水,西药组大鼠灌服百忧解,中药组大鼠灌服中汤剂柴胡龙骨牡蛎汤,连续给药28 d。分别记录各组大鼠敞箱试验评分和游泳不动时间。给药结束后测量大鼠左、右心室质量、体质量、室间隔厚度,计算大鼠左、右心室质量与体质量比值;高效液相色谱法检测大鼠脑组织5-羟色胺(5-HT)含量;酶联免疫法检测大鼠血浆血管紧张素-Ⅱ(AgⅡ)和醛固酮(ALD)含量。结果:模型组大鼠的旷场试验评分和脑组织中5-HT含量均低于正常组,游泳不动时间长于正常组,血浆AgⅡ和ALD含量、左心室质量/体质量、右心室质量/体质量、室间隔厚度均高于正常组(P <0. 05)。西药组和中药组大鼠敞箱试验评分和脑组织中5-HT含量高于模型组,左心室质量/体质量、右心室质量/体质量、室间隔厚度、游泳不动时间、血浆AgⅡ和ALD含量低于模型组(P <0. 05);西药组和中药组之间无显著差异。结论:柴胡龙骨牡蛎汤可有效改善心肌梗死后并发抑郁大鼠的抑郁症状,并且能够逆转心肌梗死大鼠的心肌重塑。
        Objective: To observe the pharmacodynamic actions of Chaihu Longgu Muli Tang on rats of depression after myocardial infarction. Methods: 68 rats were divided into 4 groups randomly: normal group,model group,Chinese medicine group and Western medicine group. Rats were made myocardial infarction model and depression model successively except for normal group. The rats of normal and model group were given 0. 9% sodium chloride solution. The rats in Chinese medicine group were given Chaihu Longgu Muli Tang and the rats of Western medicine group were given Baiyoujie for 28 days. Open-field scores and swimming no-moving time were tested. The ratio of left-ventricular to body weight and right-ventricular to body weight were calculated and the thickness of ventricular was measured. High performance liquid chromatography( HPLC) method was used to test the levels of 5-HT in brain,and the levels of plasma angiotensin Ⅱ( AgⅡ) and aldosterone( ALD) were measured with enzyme-linked immunosorbent assay( ELISA) methods. Results: Compared with normal group,open-field scores and 5-HT content in brain of model rats were reduced,swimming no-moving time was prolonged significantly,ratio of left-ventricular to body weight and right-ventricular to body weight were raised and plasma AgⅡ and ALD levels were increased. To compare with the model group,open-field scores and 5-HT content in brain of Chinese and Western medicine groups rats were increased,swimming no-moving time was shortened,ratio of left-ventricular to body weight and right-ventricular to body weight were decreased and plasma AgⅡ and ALD levels were lowered,but there were no significant difference between Chinese and Western medicine groups. Conclusion: Chaihu Longgu Muli Tang can effectively improve symptoms of depression after myocardial infarction rats,and reverse rats' ventricular remodeling.
