摘要
目的探讨双氢青蒿素对人肺腺癌细胞A549凋亡及增殖影响。方法将对数生长期人肺腺癌细胞A549随机分为实验组和对照组,实验组在细胞培养液中加入500nmol/L双氢青蒿素,对照组加入二甲基亚砜,每组细胞均培养72h,采用四甲基偶氮唑盐法检测细胞增殖能力,计算增殖抑制率,采用试剂盒检测细胞半胱氨酸蛋白酶3(Caspase3)活性,采用免疫蛋白印迹法检测Bcl-2、Bax、Akt蛋白表达水平。结果实验组细胞增殖抑制率和Caspase3活性高于对照组(P<0.05);实验组Bcl-2、Akt蛋白水平低于对照组(P<0.05),Bax蛋白水平高于对照组(P<0.05)。结论双氢青蒿素可抑制人肺腺癌细胞A549增殖能力,可能通过P13K/Akt信号传导通路促进A549细胞凋亡。
        Objective To investigate the effects of dihydroartemisinin(DHA)on apoptosis and proliferation of human lung adenocarcinoma cell line A549.Methods Human lung adenocarcinoma A549 cell line in logarithmic growth phase were randomly divided into experimental group and control group.DHA with concentration of 500 nmol/L was added into cell culture in experimental group,and dimethyl sulfoxide was added in control group.Both groups were cultured for 72 h.The cell proliferation was detected by using tetramethylazozolium salt method,the activity of Caspase3 was detected by using kits,and the levels of Bcl-2,Bax and Akt protein were detected by using western blot method.Results The cell proliferation inhibition rate and Caspase3 activity in experimental group were significantly higher than control group(P<0.05).The levels of Bcl-2 and Akt protein in experimental group were significantly lower than control group(P<0.05),but level of Bax protein was significantly higher than control group(P<0.05).Conclusion DHA could inhibit the proliferation of human lung adenocarcinoma cell line A549 and promote apoptosis of A549 cells through P13 K/Akt signaling pathway.
