摘要
采用浊点滴定法测定了乙醇-K2HPO4双水相体系的双节线相图,并使用经验方程对数据进行拟合,建立了乙醇-K2HPO4双水相体系萃取螺旋霉素的方法,系统考察了K2HPO4质量分数、乙醇质量分数、螺旋霉素初始浓度、萃取温度和体系pH对分配系数和萃取率的影响。结果表明:双水相萃取螺旋霉素是自发进行、吸热熵增的过程。当K2HPO4质量分数为总体系的20%、乙醇质量分数为总体系的16%、萃取温度为25℃、体系pH=9.5时,分配系数达到47.52,萃取率达到97.97%。
        Binodal curve of ethanol-K2 HPO4 aqueous two-phase system(ATPS) was measured by the turbidimetric titration method. Empirical equations were used to correlate the bimodal curves. A method for separation of spiramycin by ethanol-K2 HPO4 aqueous two-phase system extraction was established. Parameters affecting the partition coefficient and extraction efficiency such as mass fraction of K2 HPO4, mass fraction of ethanol, initial concentration of spiramycin, extraction temperature and pH of the system were investigated. The results demonstrated that the extraction of spiramycin was a spontaneous, endothermic and entropy increasing process. When the K2 HPO4 mass fraction was 20% of the total system, the ethanol mass fraction was 16% of the total system, the extraction temperature was 25 ℃, and the pH of the system was 9.5, the partition coefficient and extraction efficiency of spiramycin reached 47.52 and 97.97%, respectively.
