摘要
目的研究盐酸左旋咪唑对免疫功能低下小鼠免疫功能的影响。方法用氢化可的松注射液肌内注射制造免疫功能低下小鼠模型(正常组除外)。按照体重将雌性BALB/c小鼠分成5组:正常组、模型组和低、中、高3个剂量实验组(盐酸左旋咪唑10,20,40 mg·kg~(-1)),灌胃给药28 d,每组10只。以乳酸脱氢酶释放法分析NK细胞活性,以分光光度法检测血清溶血素并计算半数溶血值(HC50,以OD值表示),以巨噬细胞吞噬实验检测吞噬功能并计算吞噬指数。结果正常组、模型组和低、中、高3个剂量实验组的NK细胞活性分别为1. 28±0. 27,1. 02±0. 20,1. 21±0. 27,1. 25±0. 27和1. 33±0. 22;这5组的HC50分别为29. 18±6. 54,24. 08±7. 16,32. 69±5. 85,35. 75±6. 12和38. 21±9. 16;这5组的吞噬指数分别为2. 26±0. 38,1. 34±0. 36,1. 72±0. 05,1. 84±0. 33和2. 10±0. 45;模型组与正常组相比,NK细胞活性和吞噬指数的差异均有统计学意义(均P <0. 05);低、中、高3个剂量实验组与模型组比较,NK细胞活性(低剂量实验组除外)、HC50和吞噬指数的差异均有统计学意义(P <0. 05,P <0. 01)。结论盐酸左旋咪唑有增强免疫低下小鼠免疫功能的作用。
        Objective To study the influences of levamisole hydrochloride on immune function in immunocompromised mice. Methods Hydrocortisone was given to establish immunosuppressive mouse models by intramuscular injection. Female BALB/c mice were divided into five groups: normal group,model group,and experimental groups which were given low,medium and high doses( 10,20,40 mg·kg~(-1)) of levamisole hydrochloride by intragastric administration,respectively,conti-nuously for 28 d. Each group had 10 mice. The activity of nature killer( NK)cells was analyzed by lactate dehydrogenase release assay. Serum hemolysin was measured by spectrophotometry and half hemolysis value( HC50,OD value) was calculated. Phagocytic function was measured by macrophage phagocytosis test and phagocytic index was calculated.Results NK cell activity of normal group,model group and low,medium and high-dose experimental groups were 1. 28 ± 0. 27,1. 02 ± 0. 20,1. 21 ± 0. 27,1. 25 ± 0. 27,1. 33 ± 0. 22,respectively. HC50 in the 5 groups were 29. 18 ± 6. 54,24. 08 ± 7. 16,32. 69 ± 5. 85,35. 75 ± 6. 12,38. 21 ± 9. 16,respectively. The phagocytic index in the 5 groups were2. 26 ± 0. 38,1. 34 ± 0. 36,1. 72 ± 0. 05,1. 84 ± 0. 33,2. 10 ± 0. 45,respectively. Comparison between model group and normal group,the difference of the factors( except HC50) were significant( all P < 0. 05); comparison between three doses experimental groups and model group,the difference of the factors( except low dose experimental group of NK cell activity) were significant( P < 0. 05,P < 0. 01). Conclusion Levamisole hydrochloride can enhance the immune function of immunocompromised mice.
