摘要
目的:建立双波长HPLC测定诃子中莽草酸、没食子酸和鞣花酸含量的方法。方法:采用Agilent C18柱(250mm×4.6mm,5μm),以甲醇-0.1%磷酸为流动相梯度洗脱,流速0.8m L/min,检测波长214nm(莽草酸)和270nm(没食子酸和鞣花酸),柱温25℃。结果:莽草酸、没食子酸、鞣花酸分别在各自进样量范围内与峰面积呈良好线性关系,相关系数均在0.9992以上;各成分的平均回收率均在97.1%-97.5%(n=6),RSD均小于2.0%。结论:该方法快速、准确、简便,可用于诃子中莽草酸、没食子酸及鞣花酸的含量测定。
Objective:To establish method for simultaneous determination of shikimic acid, gallic acid and ellagic acid of Fructus Chebulae by dual-wavelength HPLC.Methods:An Agilent C18 column(250mm×4.6mm, 5μm) was used with a mobile phase of methanol-0.1% phosphoric acid at 0.8m L/min by gradient elution, and the detection wavelengths were set at 214nm(shikimic acid) and 270nm(gallic acid and ellagic acid) with the column temperature of 25℃.Results:Shikimic acid, gallic acid and ellagic acid had good linear relation with peak area respectively in the concentration ranges oftheir individual sample size(r>0.999 2), and the overall average recovery rates were in the range of 95.0%-100.5%(n=6) with RSD less than 2.0%.Conclusion:The results indicated that this dual-wavelength HPLC method is rapid, accurate and simple, which can be used for the determination of shikimic acid, gallic acid and ellagic acid in Fructus Chebulae.
