摘要
目的:检测毛蕊异黄酮对肺腺癌细胞生长转移的影响。方法:将不同浓度的毛蕊异黄酮分别作用于人肺癌细胞系A549细胞,通过MTT实验检测细胞增殖情况,通过transwell实验检测细胞转移情况。利用western blot及RT-PCR的方法检测毛蕊异黄酮对A549细胞相关蛋白的作用。结果:毛蕊异黄酮可以抑制A549细胞增殖与转移功能。(25、 50、100μg/mL)毛蕊异黄酮处理A549细胞24 h后,对其增殖的抑制率分别为(39.51±0.75)%,(48.91±3.64)%和(56.49±1.54)%。25μg/mL与50μg/mL毛蕊异黄酮作用A549细胞48 h后对其转移的抑制率分别为(17.76±3.82)%和(38.71±8.23)%。毛蕊异黄酮以剂量依赖的方式抑制A549细胞中CyclinD1与MMP2蛋白的表达。结论:毛蕊异黄酮可以通过抑制CyclinD1与MMP2蛋白的表达来抑制肺癌细胞A549细胞的增殖与转移。
        Objective:To detect the effect of calycosin on the growth and metastasis of lung cancer cells.Methods:Different concentrations of calycosin were used to human lung cancer cell line A549 cells.The cell proliferation was detected by MTT assay.The cell metastasis was detected by Transwell assay.Using Western blot and RT-PCR were used to detect the effect of calycosin on A549 cells associated proteins.Results:Calycosin could inhibit the proliferation and metastasis of A549 cells(25,50,100 μg/mL)of calycosin were treated 24 hours on lung cells.The inhibition of proliferation was(39.51±0.75)%,(48.91±3.64)% and(56.49±1.54)%.The concentrations of 25 μg/mL and 50 μg/mL of calycosin were treated 24 hours on lung cells.The inhibition of migration was(17.76±3.82)% and(38.71±8.23)%.At the same time,calycosin can significantly inhibit the expressions of CyclinD1 and MMP2 in a dose-dependent manner.Conclusion:Calycosin can inhibit the proliferation and metastasis of lung cancer cell A549 cells by inhibiting the expression of CyclinD1 and MMP2.
