摘要
目的建立一种高效胃癌原位模型。方法采用慢病毒感染方法构建稳定表达荧光素酶及GFP的双标细胞系SGC-7901/FLUC/GFP。将细胞悬液注射入造模小鼠胃浆膜下层,建模后第3天通过生物发光成像观测建模情况。2周后处死小鼠,通过HE及冰冻切片观察移植瘤形态。结果 SGC-7901/FLUC/GFP双标细胞系构建成功,该稳定转染细胞系高表达荧光素酶及GFP,细胞数目与荧光及生物发光信号呈线性正相关(R2=0.9667,r2=0.9110),可用于肿瘤在体观测。手术后第3天通过生物发光实验证实模型构建成功,肿瘤位于胃区生长。2周后通过HE及冰冻切片可见异型肿瘤细胞生长,符合胃癌组织学特征。结论成功建立了简便、安全、高效的大规模胃癌原位模型,该方法便于普及,可为胃癌研究提供宝贵的研究平台和实验工具。
        Objective To establish a novel orthotopic gastric cancer model with high efficiency. Methods The human gastric cancer cell line SGC-7901 was transfected to stable expression of luciferase and GFP by lentivirus transduction(SGC-7901/FLUC/GFP). The model was developed by orthotopic transplantation of SGC-7901/FLUC/GFP into the sub-serosal layer of the stomach of nude mice. Bioluminescence imaging was used to track and monitor the tumor growth on the 3 th day. Two weeks later,8 mice were sacrificed and tumors were excised for histological identification.Results SGC-7901/FLUC/GFP was high expression luciferase and GFP. Positive correlation between cell number and bioluminescence/fluorescence intensity was observed(R2= 0. 9667,r2= 0. 9110). The orthotopic model was successfully established. On the 3 th day,bioluminescence imaging showed the tumor growth at stomach region. The HE and frozen section also confirmed the tumor growth. This method showed high efficiency with easy procedure and less mice mortality. Conclusion This method is an easy,safe and high efficient way for the establishment of orthotopic gastric cancer model on the large scale,which would provide a valuable platform for gastric cancer research.
