真武汤对扩张型心肌病大鼠心功能及RAAS的干预研究

摘要
目的:观察真武汤对扩张型心肌病大鼠的干预作用,并探讨其作用机制。方法:35只SD大鼠腹腔注射阿霉素建立扩张型心肌病模型,另10只正常大鼠为正常组,造模后将存活的扩张型心肌病大鼠随机分为培哚普利组、真武汤组、模型组,连续灌胃给药8周。对上述各组大鼠行心脏超声检查;采血测定大鼠血浆B型尿钠肽,血管紧张素Ⅱ(AngⅡ)水平,取大鼠左心室心肌进行HE染色;测定心肌组织蛋白激酶C(PKC)含量;采用RT-PCR测定心肌组织血管紧张素Ⅱ1型受体(AT1)的mRNA表达水平。结果:真武汤组可以改善扩张型心肌病模型大鼠的心腔大小、心功能、心肌损伤。真武汤组血浆AngⅡ水平、心肌组织的PKC含量、AT1的mRNA水平均低于模型组。结论:真武汤可能是通过调节血浆AngⅡ浓度、心肌组织PKC含量及AT1 mRNA表达水平,抑制RAAS激活,改善心功能。
Objective:To investigate the effects of Zhenwu decoction on dilated cardiomyopathy and explore its mechanisms.Methods:A total of 35 SPF male SD rats were used to establish dilated cardiomyopathy model(DCM) by intraperitoneal injection of Adriamycin.Survived DCM rats after modeling were randomly divided into perindopril group,Zhenwu decoction group,and model group,and fed for 8 weeks.The other 10 normal rats served as the normal group.Echocardiography was performed on the rats of all groups.Then,animals were killed to measure brain natriuretic peptide(BNP) and angiotensin Ⅱ(AngⅡ) in blood plasma,and the change of left ventricular cardiac was histological observed through hematoxylin and eosin staining.Detect the content of protein kinase C(PKC) in left ventricular myocardium.The mRNA expression levels of angiotensin type 1 receptor(AT1) in myocardium from each group were detected by reverse transcription-polymerase chain reaction(RTPCR).Results:Compared to the model group,Zhenwu decoction can improve the cardiac cavities,cardiac function,and myocardial injury in DCM rats.Compared to the model group,the plasma levels of AngⅡ,the content of PKC in left ventricular myocardium and mRNA levels of AT1 in the myocardium of Zhenwu decoction group decreased.Conclusion:Zhenwu decoction can effectively improve heart function of dilated cardiomyopathy.The mechanisms of Zhenwu decoction on DCM maybe inhibit RAAS activation by regulating plasma Ang Ⅱ concentration,PKC content in left ventricular myocardium and AT1 mRNA expression in myocardium.
