中药肾毒宁颗粒对慢性肾衰大鼠的肾保护作用及其机制研究

摘要
目的:观察肾毒宁颗粒对2/3肾切除大鼠的肾保护作用及其机制。方法:将SD大鼠120只随机分为手术组(100只)和假手术组(20只),手术组大鼠行2/3肾切除术。术后2周将手术组随机分为西药组、模型对照组及肾毒宁颗粒低、中、高剂量组,每组各18只。治疗2个月后记录比较各组血肌酐(SCr)、尿素氮(BUN)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果:与假手术组相比,模型对照组SCr、BUN、MDA水平显著升高(P<0.01),SOD水平显著下降(P<0.01)。与模型对照组相比,各治疗组SCr、BUN、MDA水平均显著降低(P<0.01),SOD水平均显著升高(P<0.01),且呈剂量依赖关系。高剂量组与西药组SOD、MDA水平比较无统计学差异(P>0.05)。结论:中药肾毒宁颗粒能够有效改善CRF大鼠肾功能,其机理可能与其通过抗氧化作用,调节SOD、MDA水平有关。
        Objective:To observe protective effect of Shendunning granule on 2/3 nephrectomized rats and its mechanism.Methods:Divided 120 SD rat into operation group(100 cases)and sham group(20 cases)randomly,and provided 2/3 nephrectomy to operation group.After 2 weeks of operation,divided operation groups into western medicine group,model control group,shenduning high,medium and low dose group randomLy with 18 cases in each group.Recorded and compared each group's serum creatinine(SCr),urea nitrogen(BUN),superoxide dismutase(SOD)and malondialdehyde(MDA)levels after 2 months' treatment.Results:Comparing with those in sham group,model control group's SCr,BUN and MDA levels increased significantly(P<0.01),but SOD decreased significantly(P<0.01).Comparing with those in model control group,each treatment group's SCr,BUN and MDA decreased significantly(P<0.01),but SOD increased significantly(P<0.01),and it is in a dose-dependent manner.Here were no significant differences if comparing SOD,MDA between shenduning high dose group and western medicine group(P>0.05).Conclusion:TCM shenduning granule helps rats with CRF to improve renal function,the mechanism maybe relates to its antioxidant to regulate SOD,MDA levels.
