摘要
目的观察千金子制霜前后石油醚提取物对大鼠十二指肠消化间期复合肌电(IMC)活动及组织匀浆P物质(SP)和血管活性肠肽(VIP)含量的影响,探讨制霜缓解千金子毒性的机制。方法将24只SD大鼠随机分为空白组、千金子生品石油醚提取物组和千金子霜品石油醚提取物组,每组8只。各给药组给予相应剂量(10 g原药材/kg)药液灌胃,空白组给予等量生理盐水灌胃。采用BL-420F生物机能实验系统测定千金子制霜前后大鼠十二指肠IMC慢波频率、振幅及峰电位数目;ELISA检测大鼠十二指肠组织匀浆SP和VIP含量。结果与空白组比较,千金子制霜前后均能兴奋大鼠胃肠肌电活动,十二指肠组织匀浆SP和VIP含量明显降低(P<0.01),制霜后各指标强度均小于生品(P<0.01)。结论千金子可能通过影响大鼠SP、VIP的分泌,进而影响消化道平滑肌胃肠电变化,引起胃肠道平滑肌收缩/舒张失衡,肠黏膜免疫调节紊乱,而制霜后使其水平趋于正常,从而改善胃肠道动力及胃肠道刺激性。
        Objective To investigate the effects of Euphorbiae Semen and Euphorbiae Semen Pulveratum extracts on interdigestive myoelectric complex(IMC) activity, the tissue content of substance P(SP) and vasoactive intestinal peptide(VIP) in rats; To explore the mechanism by which frosting can alleviate the toxicity of Euphorbiae Semen.Methods Totally twenty-four SD rats were randomly divided into control group, Euphorbiae Semen Pulveratun extracts group and Euphorbiae Semen Pulveratum extracts group. Each administration group was given relevant medicine(10 medicine/kg) for gavage, while the control group was given the same amount of saline for gavage. The BL-420 F bio-functional experimental system was used to determine the slow wave frequency, amplitude, and peak potential of rats received Euphorbiae Semen before and after frosting; the contents of duodenal tissue SP and VIP were determined by ELISA. Results Compared with the control group, Euphorbiae Semen before and after frosting could stimulate the gastrointestinal muscle activity, and reduce the contents of SP and VIP of duodenum(P<0.01).However, the intensity of Euphorbiae Semen after frosting was less than that of Euphorbiae Semen before frosting(P<0.01). Conclusion Euphorbiae Semen may affect the secretion of SP and VIP and then change gastrointestinal electrical properties of the smooth muscle of the digestive tract, causing gastrointestinal smooth muscle contraction/diastolic imbalance, local intestinal mucosal immunity regulation disorder, which can make its level tend to normal after frosting, thereby improving gastrointestinal motility and gastrointestinal irritation.
