探讨线栓法建立大鼠局灶性脑缺血模型的影响因素

摘要
目的:为了提高线栓法制备大鼠局灶性脑缺血模型成功率,探讨大鼠局灶性脑缺血模型的有效方法及影响因素。方法:为观察高温外环境对造模的影响,将外环境温度控制在(25±1)℃和在夏季未控制外环境温度(30℃以上)分为温度控制组(n=15)和非温度控制组(n=15),其他制作过程一致;为观察栓线从颈总动脉和颈外动脉插入对造模的影响分为颈总动脉插入组(n=15)和颈外动脉插入组(n=15),其他制作过程一致;为观察术中术后2h内老鼠苏醒活跃状态对造模的影响分为麻醉状态组(n=15)和苏醒活跃状态组(n=15),其他制作过程一致。从造模24h后观察Zea-Longa评分在1~3分的大鼠且7d内观察大鼠未死亡视为造模成功。结果:在外环境温度控制在(25±1)℃组与夏季未控制外环境温度(30℃以上)比较,差异无统计学意义(P=0. 107> 0. 05);线栓从颈总动脉插入比从颈外动脉插入存活率高,具有统计学意义(P=0. 025 <0. 05);术后2h内大鼠处于苏醒活跃状态组死亡率高,与麻醉未清醒状态组比较差异具有统计学意义(P=0. 021 <0. 05)。结论:直接分离出颈总动脉(CCA)并从颈总动脉插入具有易操作、创伤性小、操作时间快、进线栓快、进线栓顺利以及模型稳定等优点,能降低造模的死亡数据;术中术后2h内大鼠处于苏醒活跃状态会对模型大鼠造成较大的刺激,造成死亡率增加。
        Objective: To improve the success rate of the rat model of focal cerebral ischemia by thread embolization,and to explore the effective methods and influencing factors of the rat model of focal cerebral ischemia. Methods: In order to observe the effect of high-temperature external environment on the modeling,the external environment temperature was controlled at(25 ± 1) ℃ and uncontrolled external environment temperature in summer(over 30℃) were divided into temperature control group(n = 15) and non-temperature control group(n = 15),and the other manufacturing processes were the same. The effects of common carotid artery insertion(n = 15) and external carotid artery insertion(n = 15) were the same in the other groups,and anesthesia group(n = 15) and awakening active group(n = 15) were the same in the other groups. The rats with ZeaLonga score ranging from 1 to 3 were observed 24 hours after modeling,and the rats without death within 7 days were regarded as successful modeling. Results: There was no significant difference between the control group and the control group in summer(P = 0. 107 > 0. 05). The survival rate of the emboli inserted from the common carotid artery was higher than that inserted from the external carotid artery(P = 0. 025 < 0. 05),which was statistically significant. The rats were awake within 2 hours after operation. The mortality of active state group was higher than that of anesthesia state group(P = 0. 021 < 0. 05). Conclusion: Direct isolation of common carotid artery(CCA) and insertion of CCA from the common carotid artery have the advantages of easy operation,less trauma,fast operation time,fast thread insertion,smooth thread insertion and stable model,which can reduce the mortality data of the model. The rats in the state of awakening and activation within 2 hours after operation will cause greater stabbing in the model rats. Stimulation results in increased mortality.
