摘要
【目的】研究甘加型藏绵羊发情周期下丘脑促性腺激素释放激素(GnRH)分泌的动态变化及其对藏羊生殖活动的调控.【方法】选取发情周期不同阶段的甘加型藏绵羊48只,采集藏羊发情期、发情后期、间情期、发情前期的血浆.应用酶联免疫吸附试验(ELISA)测定各个时期血浆中GnRH的含量,比较研究其动态变化规律.【结果】甘加藏绵羊发情周期不同阶段血浆中GnRH分泌呈现一定的规律:在发情期和发情后期呈脉冲式分泌,间情期和发情前期既有脉冲式又有波动式分泌.分泌特点为发情前期血浆中GnRH(57.409pg/mL)开始升高,至达到最大值(73.389pg/mL),发情后期(62.058pg/mL)和间情期(55.687pg/mL)分泌量降低,其中发情期分泌量最多,且与其他3个时期差异显著(P<0.05).在整个发情周期中GnRH先后出现4个明显分泌峰,第1个峰出现在发情期4h,第2个峰出现在发情后期60h,第1个峰略高于第2个;第3个峰出现在间情期第9天早上,第4个峰出现在发情前期第14天早上,第3个峰较第4个低.【结论】了解甘加型藏绵羊发情周期下丘脑GnRH分泌的动态变化规律,以及GnRH在甘加型藏绵羊发情周期中的调控作用.
        【Objective】In order to study the amount of secretion changes dynamically of the hypothalamus conadotropin-releasing hormone(GnRH)of Ganjia Tibetan sheep at the estrus cycle and the relationship between GnRH and the reproductive activities of Ganjia Tibetan sheep.【Method】There were 48 sheep at different stages of the oestrous cycle,collecting the plasma of oestrus,metestrus,diestrus,proestrus of Ganjia Tibetan sheep.The content of GnRH at different stages of the oestrous cycle was determined by enzyme immune-assay(ELISA)and the dynamic variation rule was studied.【Result】The secretion of GnRH in plasma had certain regularity.The secretion of GnRH was pulsatile at the whole oestrous cycle,the sametime,the diestrus and proestrus was volatility.The characteristic of secretion was the period of estrus increased to the maximum(73.389 pg/mL),the secretion of GnRH reduced in metestrus(62.058 pg/mL)and diestrus(55.687 pg/mL),last the raised in this proestrus(57.409 pg/mL).The secretion volume of GnRH in estrus was the highest and was different from the other three periods(P<0.05).There were four secretory peaks appeared of GnRH in estrous cycle,the 1 st secretory peaks appeared in the estrus 4 h,the2 nd secretory peaks appeared in the metestrus 60 h.The 1 st secretory peak was higher than the 2 nd(P>0.05).The 3 rd secretory peaks appeared in the diestrus 9 th day,the 4 th secretory peaks appeared in the proestrus 14 th day.The 3 rd secretory was lower than the 4 th(P<0.05).【Conclusion】The dynamic changes of secretion of hypothalamus GnRH is revealed,and the regulatory role of GnRH is understood during the estrus cycle of Ganjia Tibetan sheep.
