摘要
为建立矾根组培快繁体系,以矾根品种‘花毯’的花序轴为外植体,本研究对其组培快速繁殖进行了系统研究。结果显示,适宜矾根‘花毯’建立无菌系的接种时间为4月上旬;初代诱导培养基为MS+0.6 mg/L6-BA+0.02 mg/L IBA;继代培养基为MS+0.5 mg/L 6-BA,增殖系数达9.1倍;生根培养基为MS+1.0 mg/L IBA,生根率98.05%;生根苗移栽驯化基质为泥炭土:蛭石体积比3:1,成活率达85.11%。本研究中发现,矾根以MS基础培养基添加1.0 mg/L和2.0 mg/L IBA的生根培养基中,根长、根数和生根时间差异均不显著。这说明矾根自身分化能力较强,生根培养所需外源激素较低,从节约成本角度考虑,添加一定量的MS培养基可作为‘花毯’组培苗最佳生根培养基。本研究为建立矾根组培快繁体系提供理论依据。
In order to establish a rapid propagation system for Heuchera micrantha,, the inflorescence axis of the cultivar 'Tapestry' was used as an explant. This study systematically studied the rapid propagation of its tissue culture and rapid propagation. The results showed that the inoculation time for the establishment of a sterile line for Heuchera micrantha 'Tapestry' was in early April. The initial induction medium was MS +0.6 mg/L 6-BA +0.02 mg/L IBA. The subculture medium was MS +0.5 mg/L 6-BA, and the proliferation coefficient reached by 9.1 times. Rooting medium was MS +1.0 mg/L IBA, with the rooting rate of 98.05%. Rooting shoots transplanted and domesticated matrix peat soil: vermiculite volume ratio 3:1, and the survival rate was 85.11%. In this study, the root length, number of roots, and rooting time of roots in 1.0 mg/L IBA and 2.0 mg/L supplemented with MS basic medium were not significantly different. This indicated that Heuchera micrantha had a strong ability to differentiate itself, and the exogenous hormones need for rooting culture was relatively low. From the perspective of cost saving, adding a certain amount of MS medium could be used as the best rooting medium for the 'Tapestry' tissue culture seedlings. This study could provide a theoretical basis for the establishment of a tissue culture and rapid propagation system of Heuchera micrantha.
