甲状腺癌组织中Skp2和Cx43的表达与患者临床病理特征的关系

摘要
目的研究S期激酶相关蛋白2(Skp2)和细胞缝隙连接蛋白43(Cx43)在甲状腺癌组织中的表达,分析二者相关性。方法收集2012年3月至2015年8月42例甲状腺癌组织石蜡标本和20例甲状腺肿组织标本,采用免疫组织化学S-P法检测甲状腺癌组织中Skp2、Cx43的表达,数据均纳入SPSS20.0软件分析。Skp2、Cx43蛋白阳性表达率采用例(%)表示,行χ~2检验; Skp2与Cx43相关性分析采用Spearman检验。P<0.05差异有统计学意义。结果甲状腺肿组织Skp2蛋白阳性表达率25.0%明显低于甲状腺癌组织75.0%(χ~2=8.45,P<0.05);甲状腺肿组织Cx43蛋白阳性表达率95.0%明显高于甲状腺癌组织47.6%(χ~2=9.65,P<0.05);单因素分析显示,病理分期、淋巴结转移、周围组织侵犯是甲状腺癌组织中Skp2与Cx43蛋白阳性表达率的影响因素(P<0.05);多因素分析显示,Skp2、Cx43阳性表达与肿瘤分期Ⅲ、Ⅳ期、有淋巴结转移和周围组织侵犯等因素有明显相关性(P<0.05);经Spearman相关性分析显示:Skp2、Cx43蛋白在甲状腺癌中表达呈负相关(r=-0.709,P<0.05)。结论 Skp2和Cx43均可能参与甲状腺癌的增殖、浸润及转移。Skp2、Cx43的表达情况可能作为预测甲状腺癌的恶性程度和预后的指标。
        Objective To investigate the expression of S kinase associated protein 2(Skp2) and connexin 43(Cx43) in thyroid carcinoma, and explore the correlation with their clinicopathological characteristics. Methods From March 2012 to August 2015, 42 paraffin of thyroid carcinoma, and 20 goiter tissues were collected. Immunohistochemical S-P method was used to detect the expression of Skp2, Cx43 in thyroid carcinoma. The data were analyzed by SPSS20.0 software. The positive expression rate of Skp2, Cx43 protein was expressed as n(%), and compared with χ~2 test. The correlation between Skp2 and Cx43 was analyzed by Spearman test. P<0.05 was statistically significant. Results The positive expression rate of Skp2 protein in goiter(25.0%) was significantly lower than that in thyroid carcinoma(75.0%), The difference was significant(χ~2=8.45, P<0.05); The positive expression rate of Cx43 protein in goiter(95.0%) was significantly higher than that in thyroid carcinoma(47.6%). The difference was significant(χ~2=9.65, P<0.05); Univariate analysis showed that pathological stage, lymph node metastasis and peripheral tissue invasion were the factors influencing the expression of Skp2 and Cx43 protein in thyroid carcinoma(P<0.05). Multivariate analysis showed that the positive expression of Skp2 and Cx43 was significantly correlated with the stage Ⅲ and Ⅳ of tumor, lymph node metastasis and peripheral tissue invasion(P<0.05).Spearman correlation analysis showed that: there was a negative correlation between Skp2 and Cx43 protein expression in thyroid carcinoma(r=-0.709, P<0.05). Conclusion Both Skp2 and Cx43 can participate in the proliferation,invasion and metastasis of thyroid carcinoma. The expression of Skp2 and Cx43 may be used as a predictor of malignant degree and prognosis of thyroid carcinoma.
