摘要
目的:观察桃红四物汤对2型糖尿病(T2DM)模型大鼠心脏结构和功能的干预作用及对模型大鼠心肌组织NF-κB信号通路相关分子表达的影响。方法:采用高糖高脂饲料联合链脲佐菌素制备T2DM模型大鼠,造模成功后,药物连续干预8周,观察各组大鼠心脏血流动力学及心肌组织形态学变化,同时检测大鼠心肌组织NF-κBp65、TNF-α蛋白及mRNA表达和血清IL-1β、IL-6含量。结果:T2DM模型大鼠出现显著心脏结构和功能损伤。与正常组比较,模型组大鼠心肌组织NF-κBp65、TNF-α蛋白和mRNA表达显著增加(P<0.01),血清IL-1β、IL-6含量显著升高(P<0.01)。桃红四物汤低、中、高剂量组干预8周后,可显著改善T2DM模型大鼠心脏结构和功能的损伤。与模型组比较,桃红四物汤低、中、高剂量组大鼠心肌NF-κBp65、TNF-α蛋白和mRNA表达显著减少(P<0.05,P<0.01),血清IL-1β、IL-6含量显著降低(P<0.05,P<0.01)。结论:桃红四物汤可有效改善T2DM模型大鼠心脏结构和功能,其机制与调节NF-κB信号通路过度激活有关。
Objective: To observe Taohong Siwu Decoction's intervention effect on cardiac structure and function of type2 diabetes mellitus(T2 DM) model rats and its effect on molecule expression related to myocardial tissue NF-κB signal pathway.Methods: Rats were feeding with high-fat and high-sugar foods with streptozotocin i.p. Use medicine to intervene the T2 DM models continuously for 8 weeks, then observe rats' cardiac hemodynamics and myocardial histomorphological changes, and detect rats' NF-κBp65, TNF-α proteins and mRNA expression of myocardial tissue, and IL-1β, IL-6 levels in their sera. Results:T2 DM model rats got obvious damages in both cardiac structures and functions. Comparing with the normal group, the expression of NF-κBp65, TNF-α proteins and mRNA of rats of myocardial tissue were significantly increased(P<0.01), IL-1β and IL-6 levels in their serum were significantly increased(P<0.01); After the interventions of Taohong Siwu Decoction in each dosage group for 8 weeks, the model rats' cardiac structures and functions improved significantly, their NF-κBp65, TNF-α proteins and mRNA expression of myocardial tissues were significantly decreased(P<0.05, P<0.01), IL-1β and IL-6 levels in their serum were significantly decreased(P<0.05, P<0.01). Conclusion: Taohong Siwu Decoction can effectively improve the T2 DM model rats' cardiac structure and function, and the mechanism is related to its regulation of over-activated NF-κB signal pathway.
