当归不同配伍对急性血瘀大鼠血液流变学、凝血功能及心、肺病理影响

摘要
目的:通过观察急性血瘀大鼠血液流变学、凝血功能及心、肺病理变化比较当归配伍党参、干姜、红花、香附的活血作用异同。方法:采用皮下注射盐酸肾上腺素和冰水浴共同复制急性血瘀大鼠模型,通过测定全血粘度(WBV)、血浆粘度(PV)、红细胞压积(HCT)、血沉方程K值(ESRK)、红细胞聚集指数(EAI),观察当归不同配伍对急性血瘀大鼠血液流变学的影响;通过测定凝血酶原时间(PT)、活化部分凝血酶原时间(APTT)、纤维蛋白原(FIB)、凝血酶时间(TT),观察当归不同配伍对急性血瘀大鼠凝血功能的影响;通过观察心、肺病理变化,观察当归不同配伍对急性血瘀大鼠脏腑病理的影响。结果:与模型组相比,当归-党参配伍可显著降低WBV、EAI,对心肌细胞加宽、肺出血现象有明显改善;当归-干姜配伍可显著降低WBV、PV及HCT、ESRK、EAI,延长PT、TT值,对心肌细胞间距变宽,肺泡壁增厚改善优于其他配伍组;当归-红花组可显著降低WBV、EAI及HCT、ESRK,延长PT值,对心肌细胞核变圆、肺间质水肿有一定改善;当归-香附组可降低高切变率下WBV及HCT,对凝血功能无显著改善,对于心肌细胞加宽、肺间质水肿现象有一定改善。结论:当归-党参、当归-干姜、当归-红花、当归-香附配伍均可改善急性血瘀大鼠的血液流变性、凝血系统和病理损伤程度,其中当归-干姜组的作用优于其他3个配伍组。
        Objective: To compare the hemorheology,blood coagulation and cardiac and pulmonary pathological changes in acute blood stasis rats under administration of Angelica combined with Codonopsis,Zingiber officinale,Carthamii Flos and Cyperus rotundus. Methods: The subcutaneous injection of adrenaline and ice water bath were used to establish the acute blood stasis rats model. Whole blood viscosity( WBV),plasma viscosity( PV),erythrocyte hematocrit( HCT),erythrocyte sedimentation equation K( ESRK) and erythrocyte aggregation index( EAI) were observed to study the effects of Angelica on hemorheology in rats with acute blood stasis. The prothrombin time( PT),activated partial thromboplastin time( APTT) fibrinogen( FIB),thrombin time( TT) were observed to study the effects of Angelica on coagulation function in acute blood stasis rats. Also,pathology changes of hearts and lungs were observed to study the effects of Angelica on organ pathology of acute blood stasis rats. Results: Compared with the model group,WBV and EAI in Angelica combined with Codonopsis group were significantly reduced,Angelica combined with Codonopsis significantly improved the phenomenon of myocardial cell widening and pulmonary hemorrhage. WBV,PV,HCT,ESRK,EAI,prolonged PT and TT values were significantly reduced in Angelica combined with Zingiber group,while the myocardial cell spacing change wide,alveolar wall thickening was better than other combination groups. WBV,PV,HCT,ESRK and extended PT were significantly reduced in Angelica combined with Carthamii Flos group,while the nucleus became round,lung machine edema was improved. Angelica with Cyperus rotundus reduced WBV at high shear rate and HCT and did not significantly improve coagulation function,the myocardial cells widening and pulmonary interstitial edema phenomenon were certain improved. Conclusion: Angelica with Codonopsis,with Zingiber officinale,with Carthamii Flos and with Cyperus rotundus can all improve hemorheology,coagulation system and pathological damage on acute blood stasis rats,while the effect of Angelica combined with Zingiber group has stronger effect than others.
