摘要
目的:观察痤疮合剂对兔耳痤疮模型MAPK相关信号通路的影响。方法:将全部48只随机分成6组,每组8只,即空白对照组、模型对照组、中药低剂量组、中药中剂量组、中药高剂量组、清热暗疮胶囊组。中药低、中、高剂量组按等效剂量的3、6、9倍剂量给药,清热暗疮胶囊按照等效剂量给药,每日1次,连续治疗14 d。于实验第14天后对各项指标进行观察对比。结果:TNFR1、TNF-α、IL-6、IL-8、ERK、NF-κB、p38MAPK蛋白表达平均光密度值中药高、中、低剂量组均低于模型组,且高剂量组低于中剂量组,中剂量组低于低剂量组。结论:痤疮合剂能明显减低TNFR1、TNF-α、IL-8、IL-6蛋白表达水平,干预ERK、P38MAPK、NF-κB通路,从而干预了痤疮的发展过程,且随着给药剂量的增加,机制作用逐渐加强,呈良好的量效依赖关系。
        Objective:To observe the effect of acne mixture on MAPK related signal pathway in rabbit ear acne model. Methods:All 48 rats were randomly divided into 6 groups,8 rats in each group:blank control group,model control group,low-dose group of Chinese medicine,Chinese medicine dose group,high dose of Chinese medicine group,Qingre Anchuang Capsule group. Traditional Chinese medicine is low,medium and high dose group at 3,6,9 times the dose equivalent dose administration of Qingre Anchuang Capsules according to equivalent dose,1 times daily for 14 consecutive days. After fourteenth days of experiment,the indexes were observed and compared. Results:TNFR1,TNF-α,IL-6,IL-8,ERK,NF-κB,p38 MAPK protein expression average optical density value of Chinese high,medium and low dose group were lower than those in the model group,and high dose group was lower than that of the middle dose group,middle dose group is lower than the low dose group. Conclusion:Acne mixture can obviously reduce the TNFR1,TNF-α,IL-8,IL-6 protein expression level of ERK,P38 MAPK,NF-κB,intervention of B pathway,thus interfering with the development of acne,and with the increase of the dosage,the mechanism is gradually strengthened and a good dose-response relationship.
