电针疗法对围绝经期抑郁症大鼠前额叶皮质神经细胞内尼氏小体及核仁影响的定量分析

摘要
目的:观察补电针疗法对围绝经期抑郁症大鼠前额叶皮质神经细胞病理形态改变,探讨基于"肾脑相济"理论为指导的电针疗法对此模型的治疗机制。方法:60只大鼠随机分为空白组、假手术组、模型组、西药组、电针组5组,采用去势和慢性不可预见刺激结合制备围绝经期抑郁症大鼠模型。连续治疗28 d,观察大鼠糖水实验及抓力实验,取大鼠半球固定,用尼氏染色,观察前额叶皮质神经细胞及尼氏小体形态结构,定量观察前额叶皮质区,通过计数神经细胞、测量神经细胞胞浆内尼氏小体面积、积分光密度、平均黑度及核仁数目、面积、积分光密度、平均黑度观察电针对围绝经期抑郁症模型大鼠前额叶皮质神经细胞病理形态改变的影响。结果:与空白组比较,造模后各组大鼠糖水偏嗜率及抓力有所下降(P<0.05)。治疗后,电针组及西药组与模型组比较糖水偏嗜率及抓力有所提高(P<0.05);电针组及西药组与假手术组比较糖水偏嗜率有所提高(P<0.05)。治疗后,电针组及西药组与模型组比较计数神经细胞、测量神经细胞胞浆内尼氏小体面积、积分光密度、平均黑度及核仁数目、面积、积分光密度、平均黑度均有所提高(P<0.05)。结论:补电针疗法对围绝经期抑郁症大鼠前额叶皮质神经细胞尼氏小体及核仁状态产生影响,这可能是电针能够产生神经保护作用的基础。
        Objective: To observe the effect of Bushen Jiannao acupuncture therapy of perimenopausal depression rats' prefrontal cortex nerve cells in pathological study,based on the kidney-brain theory guided mechanism of electro acupuncture therapy.Methods: Sixty rats were randomly divided into control group,sham group,model group,western medicine group and electro acupuncture group. We prepared perimenopausal depression rat model by OVX and CUMS. For 28 consecutive days,we observed the changes of sugar experiment and gripped the experimental results,the half of brain tissue and carried out Nissl staining. We observed cortex nerve cells and Nissl body structure,prefrontal cortex through the nerve cell count of nerve cells,measured pulp Nissl's body area,integral density and average number,density and nucleolus area,integral optical density and the average density index to investigate the effect of electroacupuncture on perimenopausal depression model changes in prefrontal cortex nerve cells in pathological big rat. Results: After treatment,compared with the control group,western medicine group and model group's partial addicted to sugar water rate and holding power increased(P < 0. 05). Electro acupuncture group and western medicine group' s sugar water partial heavy rate increased compared with that of the control group(P < 0. 05). After treatment,the curative group and western medicine group's count of nerve cells,nerve cells in the cytoplasm of nissl body area,integral density,average number of blackness and nucleoli,integral optical density and the average black degree increased compared with those of the model group(P < 0. 05). Conclusion: Bushen Jiannao electroacupuncture therapy has an effect on the Nissl body and nucleolus of the prefrontal cortex of the rats with perimenopausal depression.
