摘要
目的建立尿中硫氰酸盐的酶标仪快速测定法。方法尿样在微酸条件下与氯胺T、吡啶反应,其反应产物与巴比妥酸反应显红色,采用酶标仪测定,并对结果进行肌酐校正。结果硫氰酸盐在40~800μg/L范围内该方法线性关系良好(r=0.999 8),检出限为1.1μg/L,平均回收率为88.9%~101.2%,RSD为5.8%~7.2%。结论该方法简便、快速、灵敏度高,适用于职业人员尿中硫氰酸盐的测定。
Objective To establish a method for rapid determination of thiocyanate in urine samples of workers with the microplate reader. Methods The urine sample reacted with chloramines T and pyridine at weak acid condition, the product from which reacted with barbituric acid and appeared red, then was tested with microplate reader, and the results were corrected with creatinine. Results The linear range was 40-800 μg/L thiocyanate and the correlation coefficient was 0.999 8.The means of recovery rate were 88.9%-101.2% and RSDs were 5.8%-7.2%(n =5).The detection limit was 1.1 μg/L.Conclusion This method is simple, rapid, accurate and is applicable to the determination of thiocyanate in the urine of population with occupational exposure.
