摘要
目的观察参慈胶囊联合放疗对人肺鳞癌细胞株SK-MES-1裸鼠移植瘤血管生成的影响,并探讨其可能的作用机制。方法在无菌条件下每只裸鼠右侧背部皮下接种单细胞悬液制备荷瘤小鼠模型,将32只BALB/c裸鼠随机分为4组。(1)模型组:予生理盐水灌胃,连续15天后断颈处死。(2)参慈胶囊组:每日予参慈胶囊提取液110 g/kg裸鼠,各组均以0. 2 m L灌胃,连续15 d后断颈处死。(3)放射组与参慈胶囊+放射组:每日予参慈胶囊提取液110 g/kg裸鼠并且在第1、3、5、8、10、12天行采用6-MV的X线照射,总剂量为18 Gy/6F。各组连续治疗15d后断颈处死。取肿瘤组织采用免疫组化SP方法检测各组肿瘤组织血管生成情况,采用荧光定量PCR方法检测各组肿瘤组织VEGF与KDRmRNA表达的情况。结果参慈胶囊配合放射治疗能够抑制人肺鳞癌细胞株SK-MES-1裸鼠移植瘤肿瘤的血管生成。结论参慈胶囊能够抑制裸鼠移植瘤血管生成,其机制可能是通过降低VEGF和KDR的表达,或同时对二者进行调控而达到增敏效应。
Objective To observe the effect of Shenci capsule combined with radiotherapy on angiogenesis of human lung squamous cell carcinoma SK-MES-1 in nude mice and explore its possible mechanism. Methods: Under sterile condition,each nude mouse was subcutaneously inoculated with a single cell suspension on the right back to prepare a tumor-bearing mouse model. BALB/c nude mice( n = 36) were randomly divided into four groups:(1)Model group: nude mice was fed normalsalinefor 15 days and guillotined at last.(2)Shenci capsule group: nude mice was fed 110 g/kg of Shenci capsule extracting solution( 0. 2 m L) for 15 days and guillotined at last.(3)Radiation group and Shenci Capsule + Radiation group:nude mice was fed 110 g/kg of Shenci Capsule extracting solution every day for 15 days. In the meantime,6-MV X-ray irradiation was performed on the 1 st,3 rd,5 th,8 th,10 th and 12 th days,the total dose was 18 Gy/6 F. All mice were guillotined after 15 days of continuous treatment. Immunohistochemical SP method was used to detect the angiogenesis of tumor tissues in each group,and quantitative fluorescence PCR was used to detect the expression of VEGF and KDR in each group.Results: Shenci capsule combined with radiotherapy could inhibit angiogenesis of SK-MES-1 transplantedtumorin nude mice. Conclusion Shenci Capsule can inhibit angiogenesis in nude mice transplanted tumors. Its mechanism may be through reducing the expression of vascular endothelial growth factor and KDR,or regulating both of them simultaneously to achieve sensitization effect.
