秦岭龙胆挥发油成分分析及抗氧化活性研究

摘要
目的对比秦岭龙胆挥发油酯化前后化学成分及相对含量的变化,并评价其抗氧化能力。方法利用超声辅助提取技术提取秦岭龙胆中的挥发油,并对挥发油进行甲酯化。利用气相色谱-质谱联用技术(GC-MS)对酯化前后的化学成分进行分析;利用清除DPPH自由基及ABTS自由基测定秦岭龙胆挥发油的抗氧化活性。结果GC-MS结果显示,秦岭龙胆挥发性成分组成在酯化前后发生了较大变化,酯化前共分析出26种成分,酯化后分析出25种成分,其中有相同成分16种;其主要成分含量在酯化前后亦发生了较大变化,如不饱和脂肪酸亚油酸从酯化前的1.56%上升到酯化后的6.72%,油酸从酯化前的0.42%上升到4.92%;秦岭龙胆总挥发油有一定的抗氧化活性,对DPPH及ABTS自由基阳离子均有一定的清除作用,其中对DPPH的清除率远大于标准品抗氧化剂VC。结论秦岭龙胆挥发性成分组成在酯化前后组成及含量均发生了较大变化,且秦岭龙胆挥发油成分有一定的抗氧化活性。
        Objective To study the change of chemical composition and relative content of volatile oil before andafter esterification in volatile oil from Gentiana apiata N. E. Br.,and evaluate its antioxidant activity. Method Thevolatile oil from Gentiana apiata was extracted by ultrasonic assisted extraction, and the volatile oil was methylesterified. The change of its components before and after esterification was examined by GC-MS. Antioxidant activitywas assessed by scavenging rate of DPPH and ABTS+·. Results The volatile oil composition changed greatly beforeand after esterification. A total of 26 components were detected before esterification, and 25 components weredetected after esterification, of which 16 were the same components. The main components' contents were alsogreatly changed,for example,unsaturated fatty acid linoleic acid increased from 1.56 % to 6.72 %,and oleic acidincreased from 0.42 % to 4.92 %. The volatile oil from Gentiana apiata could scavenge DPPH and ABTS+·,and theremoval rate of DPPH is much greater than the standard antioxidant VC. Conclusion The composition and contentof the volatile oil from Gentiana apiata changed greatly before and after esterification, and the volatile oil from Gentiana apiata showed certain antioxidant activity.
