摘要
目的:优选秦岭龙胆中齐墩果酸的乙醇回流提取工艺。方法:采用HPLC测定齐墩果酸含量,流动相乙腈-甲醇-水-磷酸-三乙胺(25∶15∶10∶0.01∶0.02),检测波长210 nm。在单因素试验基础上,根据Box-Benhnken中心组合设计原理,利用三因素三水平响应面分析法考察料液比、提取次数、提取时间对齐墩果酸提取量的影响。结果:最佳提取工艺为料液比1∶12,提取数2次,提取时间76 min;齐墩果酸提取量0.583 mg·g-1,与预测值0.605 mg·g-1的偏差仅3.64%。结论:采用响应面法优选的秦岭龙胆回流提取工艺稳定可行,为秦岭龙胆的资源开发及临床推广提供参考。
        Objective: To optimize ethanol reflux extraction process of oleanolic acid in Gentiana apiata.Method: HPLC was employed to determine the content of oleanolic acid with mobile phase of acetonitrilemethanol-water-phosphoric acid-triethylamine( 25∶ 15∶ 10∶ 0. 01∶ 0. 02) and detection wavelength at 210 nm. On the basis of single factor tests,according to Box-Behnken design principles,response surface analysis was adopted to investigate effects of solid-to-liquid ratio,extraction times and extracting time on yield of oleanolic acid. Result:Optimal extraction conditions were as follows: solid-to-liquid ratio of 1 ∶ 12,extracted twice,extracting time of76 min; under these conditions, yield of oleanolic acid was 0. 583 mg ·g- 1,closed to predicted yield of0. 605 mg·g- 1. Conclusion: This optimized process was feasible and stable,it had a certain reference value for resource development and clinical promotion of G. apiata.
