摘要
目的:研究大孔吸附树脂分离纯化积雪草中积雪草总苷的工艺条件。方法:以静态饱和吸附量、静态洗脱率为考察指标,比较了4种大孔吸附树脂分离、纯化积雪草总苷的工艺。以积雪草总苷含量、吸附率以及洗脱率为指标,对最佳树脂吸附工艺条件进行了筛选。结果:大孔吸附树脂纯化工艺为选用DPH~(-1)00型大孔树脂,上样液质量浓度为0.3 g﹒mL~(-1),上样体积60 mL,洗脱流速2 BV﹒h~(-1),乙醇洗脱体积分数50%,乙醇洗脱体积4 BV,洗脱后积雪草总苷含量高达60.67%。结论:该方法可行,重现性,可用于分离纯化积雪草中积雪草总苷
        Objective: To study the technological conditions for the separation and purification of asiaticosides by macroporous adsorption resin. Method: The static adsorption capacity and static elution rate were used as indexes to optimize the separation and purification of asiaticosides by four macroporous adsorption resins. The best technological condition for the separation and purification of asiaticosides by macroporous adsorption resin was optimized with asiaticosides content, the rates of adsorption and elution. Result: Macroporous adsorption resin purification process was as follows: DPH~(-1)00 macroporous resin, the concentration of sample solution was 0.3 g﹒mL~(-1), the sample volume was 60 mL, elution rate 2 BV﹒h~(-1), elution volume fraction of 50% ethanol, ethanol volume 4 BV. The content of asiaticosides was 60.67% after elution. Conclusion: The method is feasible and reproducible, and it can be use to separate and purify asiaticosides.
