摘要
目的:观察隐丹参酮对HTR-8/SVneo细胞增殖、凋亡、侵袭、迁移及人绒毛膜促性腺激素(hCG)表达的影响。方法:不同浓度的隐丹参酮处理细胞后,CCK8检测细胞增殖,划痕实验检测细胞迁移,transwell实验检测细胞侵袭,流式细胞仪检测细胞凋亡,ELISA检测细胞培养上清hCG表达。结果:与空白组比较,隐丹参酮5、10、20、40μmol/L作用48h可显著抑制HTR-8/SVneo细胞增殖(P<0.01),隐丹参酮20μmol/L可抑制HTR-8/SVneo细胞迁移,隐丹参酮20、40μmol/L作用24h可显著抑制HTR-8/SVneo细胞侵袭(P<0.01),隐丹参酮40μmol/L作用24h可显著诱导HTR-8/SVneo细胞凋亡(P<0.01),隐丹参酮20、40μmol/L作用24h可显著抑制HTR-8/SVneo上清β-hCG表达(P<0.05)。结论:隐丹参酮能抑制HTR-8/SVneo细胞增殖及侵袭迁移,诱导凋亡,并抑制HTR-8/SVneo细胞合成分泌hCG。
        Objective: To observe the intervention effects of cryptotanshinone on the proliferation, apoptosis, invasion,migration and human chorionic gonadotropin(hCG) expression of human chorionic trophoblast HTR-8/SVneo cells. Methods:HTR-8/SVneo cells were treated with different concentration of cryptotanshinone, cell proliferation, migration invasion and apoptosis were detected by CCK8, wound scratch assay, transwell and flow cytometry. Enzyme-Linked Immuno Sorbent Assay(ELISA) was used to detect hCG expression in cell supernatant. Results: Compared with the control group, 5, 10, 20 and40μmol/L of 48 h exposure of cryptotanshinone could significantly inhibit the proliferation of HTR-8/SVneo cells(P<0.01),20μmol/L of cryptotanshinone could inhibit the migration of HTR-8/SVneo cells, 20 and 40μmol/L of 24 h exposure of cryptotanshinone could significantly inhibit the invasion of HTR-8/SVneo cells(P<0.01). The apoptosis of HTR-8/SVneo cells was significantly induced by 40μmol/L of 24 h exposure of cryptotanshinone(P<0.01), and the levels of β-hCG in HTR-8/SVneo supernatants was significantly inhibited by 20 and 40μmol/L of 24 h exposure of cryptotanshinone(P<0.05). Conclusion:Cryptotanshinone can inhibit the proliferation, invasion, migration and hCG synthesis and secretion as well as induce apoptosis of HTR-8/SVneo cells.
