桔梗叶片愈伤组织诱导分化研究

摘要
以桔梗叶片作为外植体,采用酒精和氯化汞组合消毒,研究不同消毒时间对消毒效果的影响,并比较不同浓度激素下愈伤组织诱导及分化的变化。结果表明,桔梗外植体最适合的消毒方案为:酒精消毒30 s后氯化汞消毒6 min;桔梗叶片愈伤组织诱导的最适培养基为:1/2MS+6-BA 2.0 mg/L+NAA 0.5 mg/L;桔梗叶片诱导分化的最适培养基为:1/2MS+6-BA 0.1mg/L+NAA 0.2mg/L,此培养基培养,桔梗不定芽的诱导分化率能达到37%,且不定芽数量多,芽强壮。
        Using balloon flower(Platycodon grandiflorus) leaves as explants, the disinfection effect of different disinfection time of alcohol and mercury chloride was studied, and the changes of callus induction and differentiation under different concentrations of hormones were compared. The results showed that the most suitable disinfection scheme for Platycodon grandiflorus explants was to disinfect the explants in mercury chloride for 6 minutes after 30 seconds of alcohol disinfection. The optimum medium for callus induction of Platycodon grandiflorus was 1/2 MS+6-BA 2.0 mg/L+NAA 0.5 mg/L; the optimum induced medium for differentiation of Platycodon grandiflorus leaves was 1/2 MS+6-BA 0.1 mg/L+NAA 0.2 mg/L, then the induced differentiation rate of adventitious buds of Platycodon grandiflorus reached 37%, and the number of adventitious buds was large, and the buds were strong.
