摘要
目的建立高效液相色谱法(HPLC)同时测定石龙清血颗粒中没食子酸、原儿茶酸、马钱苷、芦丁的含量。方法采用高效液相色谱法,Kromasil C_(18)(250 mm×460 mm,5μm)色谱柱,以乙腈(A)-0.1%磷酸水(B)为流动相进行梯度洗脱,流速为1.0 mL·min~(-1),柱温为30℃,检测波长为240 nm。结果石龙清血颗粒中没食子酸、原儿茶酸、马钱苷、芦丁的线性范围分别为33.4~100.2、15.0~44.8、82.0~244.9、450.0~1343.7 mg·L~(-1),相关系数r均大于等于0.999 0,加样回收率均符合要求,含量分别为0.6577、0.2662、1.195和6.652 mg·g~(-1),含量RSD分别为0.9%、1.0%、1.4%和1.2%。阴性对照无干扰,各峰分离度良好。结论所建立的方法适用于石龙清血颗粒的质量控制。
Objective To establish a high performance liquid chromatography(HPLC) method to determinate the content in Shilongqingxue Granules(gallic acid,protocatechuic acid,loganin and rutin).Methods The column was Kromasil C_(18)(250 mm × 4.6 mm,5μm):acetonitrile(A)-0.1% phosphoric acid(B) was used as mobile phase in gradient elution:the flow rate was 1.0 mL·min~(-1):column temperature was 30 °C:the detection wavelength was 240 nm.Results The linear ranges of gallic acid,protocatechuic acid,loganin and rutin in Shilongqingxue Granule were 33.4-100.2、15.0-44.8、82.0-244.9、450.0-1 343.7 mg·L~(-1),respectively. The correlation coefficient r was greater than or equal to 0.999 0. The recovery rates of the samples were all between 95% and 105%:the contents were 0.6576,0.266 2,1.195,and 6.652 mg·g~(-1),respectively,and the RSD were 0.9%、1.0%、1.4% and 1.2%,respectively. The negative control had no interference and the peaks were well separated.Conclusion This method is applicable to the quality control of Shilongqingxue granules.
