摘要
目的:观察山姜属中药红豆蔻与大高良姜乙酸乙酯部位对胃溃疡寒证模型大鼠胃泌素(GAS)、胃动素(MOT)、表皮生长因子(EGF)、血管内皮生长因子(VEGF)、6-酮-前列腺素F1α(6-keto-PGF1α)的影响,以期探讨其对胃溃疡寒证大鼠温胃散寒的作用机制。方法:70只大鼠随机分为空白组、模型组、阳性对照组、红豆蔻与大高良姜乙酸乙酯高低剂量组(分别称为红乙高、低组;大乙高、低组),每组10只。采用15%冰乙酸和4℃知母水煎液复制胃溃疡寒证模型。造模成功后,4个受试药物组分别灌胃红豆蔻与大高良姜乙酸乙酯高、低剂量药物,测定大鼠胃组织GAS、MOT、EGF、VEGF、6-keto-PGF1α的含量。结果:与模型组比较,各给药组GAS、MOT含量显著降低(P<0.01);EGF、VEGF、6-keto-PGF1α含量显著升高(P<0.01,P<0.05,除红乙低组6-keto-PGF1α外);与大乙高组比较,红乙高组可显著降低GAS、MOT含量(P<0.01),显著升高VEGF含量(P<0.05)。结论:红豆蔻与大高良姜乙酸乙酯部位可通过降低GAS、MOT含量,升高EGF、VEGF、6-ketoPGF1α含量而发挥其温胃散寒的功效及胃溃疡保护作用。
        Objective: To study the effects and mechanisms of ethyl acetate parts of Alpinia galanga Will. and Alpinia galangal(L.) Will on GAS, MOT, EGF, VEGF and 6-keto-PGF1α contents in gastro-tissue of gastric ulcer rats with cold syndrome, and to research the pharmacodynamic mechanism of them. Methods: Seventy rats were randomly divided into 7 groups: blank group, model group, positive control group, high and low dose ethyl acetate parts of Alpinia galanga Will. groups, high and low dose ethyl acetate parts of Alpinia galangal(L.) Will. groups, 10 mice each group. Gastric ulcer rats with cold syndrome models were established by 15%glacial acetic acid and 4℃ water decoction of Rhizoma Anemarrhenae. Four TCM treated groups were administrated with corresponding medicines. GAS, MOT, EGF, VEGF, 6-keto-PGF of stomach tissues were determined. Results: Compared to the model group, each dosage group significantly reduced the concentration of stomach GAS, MOT(P<0.01) and remarkably increased EGF, VEGF, 6-keto-PGF1α(P<0.01, P<0.05), except the low does ethyl acetate parts of Alpinia galanga Will.'s 6-keto-PGF1α. Compared with the high dose group of the ethyl acetate parts of Alpinia galangal(L.)Will., the high doses of the ethyl acetate parts of Alpinia galanga Will. significantly reduced the concentration of stomach GAS, MOT(P<0.01) and increased VEGF remarkably(P<0.05). Conclusion: The ethyl acetate parts of Alpinia galanga Will. and Alpinia galangal(L.)Will. has good improving effect by decreasing the content of GAS, MOT, and increasing the content of EGF, VEGF, 6-keto-PGF1α. It is indicated that the ethyl acetate parts of Alpinia galanga Will. and Alpinia galangal(L.) Will. are the main active parts and has close relevance with the function of warming stomach for dispelling cold and protective effect for gastric ulcer.
