摘要
目的:建立大高良姜、红豆蔻、高良姜、益智、草豆蔻5味山姜属中药正丁醇部位的HPLC指纹图谱,探讨同亲缘关系中药化学成分的相关性。方法:以80%乙醇作为提取溶剂,对大高良姜、红豆蔻、高良姜、益智、草豆蔻进行加热回流提取,提取液浓缩后依次用石油醚、乙酸乙酯、水饱和正丁醇萃取,萃取液浓缩成浸膏,得到正丁醇部位,用甲醇配制成50 mg/mL供试品溶液。采用HPLC色谱法测定,国家药典委员会相似度评价软件进行分析。结果:建立了精密度、稳定性和重现性均较好的5味山姜属中药正丁醇部位HPLC指纹图谱,确定6个共有峰;其中高良姜、大高良姜含有11个共有峰,高良姜和草豆蔻含有12个共有峰,大高良姜和红豆蔻含有14个共有峰,红豆蔻、高良姜、益智和草豆蔻含有9个共有峰。大高良姜、红豆蔻、高良姜、益智、草豆蔻正丁醇部位化学成分相似度分别为0.344、0.756、0.484、0.596、0.680。结论:5味山姜属中药正丁醇部位含有共性成分较多,且含有较多极性相近的成分,红豆蔻、益智、草豆蔻正丁醇部位相似性较大,大高良姜、高良姜相似性较小。说明同亲缘关系中药化学成分有较密切的相关性。
        Objective:To establish the HPLC fingerprints of n-butanol parts from five traditional medicine[Alpinia galangal(L.)Wild.,Alpinia galanga Will.,Alpinia officinarum Hance,Alpinia oxyphyllaMiq,Alpinia galangal],and to explore the correlation of related traditional Chinese medicine on chemical composition. Methods:The five traditional Chinese medicine were extracted by 80% ethanol and four extracts of petroleum ether,ethyl acetate,n-butanol and water were obtained from ethanol extract of five traditional Chinese medicine. Methanol was used as n-butanol part solvent(50 mg/mL). HPLC method was used to analysis the five traditional medicines. The data were evaluated by using the similarity evaluation system for chromatographic fingerprint of TCM software. Results:The established HPLC fingerprint chromatogram of five traditional Chinese medicine was precise,reproducible and stable. Five traditional Chinese medicine have the six common peak. Alpinia officinarum Hance and Alpinia galangal(L.)Wild. share eleven common peak. Alpinia officinarum Hance and Alpinia galangal share twelve common peak. Alpinia galangal(L.)Wild. and Alpinia galanga Will. share fourteen common peak. Alpinia galangaWill.,Alpinia officinarum Hance,Alpinia oxyphylla Miq and Alpinia galangal share nine common peak. The similarity of chemical composition from Alpinia galangal(L.)Wild.,Alpinia galanga Will.,Alpinia officinarum Hance,Alpinia oxyphylla Miq,Alpinia galangal in n-butanol part were 0.344,0.756,0.484,0.596,0.680. Conclusion:The similarity of Alpinia galanga Will.,Alpinia oxyphylla Miq and Alpinia galangal in n-butanol parts were higher than Alpinia galangal(L.)Wild. and Alpinia officinarum Hance. There are many common components and close characters in n-butanol parts of the five traditional Chinese medicine. It illustrates chemical composition has a certain correlation between related traditional Chinese medicine.
