摘要
为了探讨黄芩苷对副猪嗜血杆菌(HPS)诱导后猪血管内皮细胞黏附分子E-选择素(E-selectin)、细胞间黏附分子1(ICAM-1)和血管细胞黏附分子(VCAM-1)的影响,体外培养猪血管内皮细胞,接种副猪嗜血杆菌。用ELISA法检测细胞培养液中E-selectin、ICAM-1和VCAM-1释放量;用Q-RT PCR法检测血管内皮细胞内E-selectin、ICAM-1和VCAM-1的mRNA表达水平。结果表明:HPS感染猪血管内皮细胞后,细胞培养液中E-selectin、ICAM-1、VCAM-1的释放量和细胞中E-selectin、ICAM-1、VCAM-1的mRNA表达水平都极显著地上调(P <0. 01);加入药物干预后,100μg/mL的黄芩苷能极显著地抑制细胞培养液中E-selectin、ICAM-1、VCAM-1的释放量和细胞中E-selectin、ICAM-1、VCAM-1的mRNA表达水平(P <0. 01)。实验表明HPS感染猪血管内皮细胞后可以激活细胞黏附分子,黄芩苷可以抑制细胞黏附分子的表达。
In order to investigate the effect of baicalin on the expression of vascular endothelial cell adhesion molecules( E-selectin,ICAM-1,VCAM-1) induced by Haemophilus parasuis( HPS),porcine vascular endothelial cells were co-cultured with Haemophilus parasuis. The amount of E-selectin,ICAM-1 and VCAM-1 in cell culture medium was detected by ELISA. The mRNA expression levels of E-selectin,ICAM-1 and VCAM-1 in vascular endothelial cells were detected by Q-RT PCR. The results showed that after HPS infection with porcine vascular endothelial cells,the release of E-selectin,ICAM-1,VCAM-1 and the expression of E-selectin,ICAM-1 and VCAM-1 at mRNA levels were significantly up-regulated( P < 0. 01); After adding drugs,baicalin at 100 μg/m L can significantly inhibit the release of E-selectin,ICAM-1,VCAM-1 in cell culture and E-selectin,ICAM-1,VCAM-1 mRNA expression levelsin vascular endothelial cells( P < 0. 01). Our results showed that HPS could induce the expression of cell adhesion molecules and baicalin could inhibit the expression of cell adhesion molecules.
