摘要
为了提高水稻小孢子离体培养中愈伤组织诱导和再生效率,以4份水稻为材料,研究了温度、渗透压(甘露醇)和化学(秋水仙碱)等预处理离体花药和小孢子对愈伤产量和绿苗分化的影响。结果表明:水稻离体花药分别于60 g/L甘露醇和10 mg/L秋水仙碱中预处理3 d均能明显提高小孢子愈伤组织产量及绿苗产量;游离小孢子于32℃热击预处理2 d也能提高小孢子愈伤组织产量和绿苗产量
        In order to improve the efficiency of callus induction and regeneration in rice( Oryza sativa) microspore culture in vitro,the in vitro anther was pretreated with mannitol and colchicine at various temperatures.The results showed that the 3-d pretreatment of the in vitro anther with 60 g/L mannitol or 10 mg/L colchicine could obviously improve the callus yield and the green plantlet yield,and the 2-d 32 ℃ heat shock pretreatment of the free microspores could also increase both callus and green plantlet yields.
