摘要
目的:观察枸杞多糖对HT22细胞缺糖缺氧再灌注损伤的影响。方法:构建HT22细胞缺糖缺氧再灌注损伤模型,设正常组、模型组、枸杞多糖100,50和25 mg·L~(-1)组,检测细胞存活率;并通过HE染色观察形态学改变;检测细胞内SOD,GSH-PX,MDA,T-AOC;利用流式细胞术检测线粒体膜电位。结果:枸杞多糖100和50 mg·L~(-1)组能明显提高细胞存活率,显著改善缺氧缺糖再灌注损伤引起的形态改变;可显著提高细胞内SOD,GSH-PX及T-AOC,减少MDA的产生;改善线粒体膜电位下降。结论:枸杞多糖能够显著减轻缺糖缺氧再灌注损伤引起的HT22细胞过氧化损伤。
        Objective: To observe the protective effects of Lycium barbarum polysaccharides( LBP) on oxygen-glucose deprivation and reperfusion in mouse hippocampal neurons. Methods: Mouse hippocampal neurons were cultured to establish oxygen-glucose deprivation and reperfusion model. Cells were assigned to control group,model group and LBP group,and the survival rate of cells was measured; morphological changes were observed by HE staining; levels of SOD,GSH-PX,MDA,and T-AOC in the cells were evaluated; the mitochondrial membrane potential were measured. Results: LBP( 100,50 mg·L~(-1)) can significantly reduce the survival rate of nerve cells,improve cell morphology,increase cell SOD,GSH-PX,T-AOC,inhibite MDA,and improve the membrane potential of mitochondria. Conclusion: LBP have significant protective effects on oxygen-glucose deprivation and reperfusion,which may be mediated by improving the antioxidant capacity of neuronal cells.
