摘要
目的 观察艾灸治疗大鼠实验性类风湿关节炎(RA)的效果及其肠道菌群变化。方法 将46只SD大鼠随机分为正常对照组10只,正常艾灸组12只,RA模型组12只,RA艾灸组12只。RA艾灸组与RA模型组以牛Ⅱ型胶原诱导方法建立关节炎大鼠模型。正常艾灸组与RA艾灸组给予艾灸双侧肾俞、足三里,正常对照组与RA模型组不进行艾灸治疗。分别于造模成功后,艾灸干预第1、2、3周测量各组大鼠体质量、足趾容积、关节炎指数(AI)评分、杆菌/球菌、革兰阳性杆菌/细菌总数并比较差异。结果 大鼠造模后较正常对照组一般情况差、体质量低、足趾容积大、AI评分高、杆菌/球菌及革兰阳性杆菌/细菌总数比值低。艾灸干预后一般情况改善、体质量增加、足趾容积稳定增加、AI评分下降、杆菌/球菌及革兰阳性杆菌/细菌总数比值增加。RA临床表现和肠道菌群变化呈显著性相关,各时间段艾灸干预后RA模型组、RA艾灸组大鼠杆菌/球菌、革兰阳性杆菌/细菌总数差异有统计学意义(P<0.05或P<0.01)。结论 艾灸能够有效治疗Ⅱ型胶原诱导关节炎大鼠关节炎症和调节肠道菌群紊乱状态。
        Objective To observe the effect of moxibustion on experimental rheumatoid arthritis(RA) and changes of intestinal flora in rats. Methods Forty-six SD rats were randomly divided into normal control group(10 rats), normal moxibustion group(12 rats), RA model group(12 rats) or RA moxibustion group(12 rats). The rats in RA model group and RA moxibustion group were treated using bovine type II collagen induction method to induce RA. The normal moxibustion group and RA moxibustion group were given moxibustion on both sides of Shenshu and Zusanli. The normal control group and RA model group were not given moxibustion treatment. The body weight, toe volume, AI score, ratios of bacillus/coccus and gram-positive bacilli/total number of bacteria were measured after successful modeling and 1, 2 and 3 weeks after moxibustion intervention. Results The clinical manifestations of rats after successful modeling were worse than those in the normal control group, with lower body weight, larger toe volume, higher AI score, and lower ratios of bacillus/coccus and gram-positive bacilli/total number of bacteria(P<0.05 or P<0.01). After moxibustion intervention, the general condition improved, body weight increased, toe volume increased steadily, AI score decreased, and the ratios of bacillus/coccus and gram-positive bacilli/total number of bacteria increased. The clinical manifestations of RA and intestinal flora were significantly correlated. There were significant differences in the ratios of bacillus/coccus and gram-positive bacilli/total number of bacteria between the RA model group and RA moxibustion group(P<0.05 or P<0.01). Conclusion Moxibustion can effectively treat arthritis rats induced by collagen type II and regulates intestinal flora disorder; the latter may be one of the important mechanisms of moxibustion in treating rheumatoid arthritis.
