冰黄五苓散直肠灌注对缺血性脑卒中大鼠神经功能的影响

摘要
目的:观察冰黄五苓散直肠灌注对缺血性脑卒中大鼠神经功能的影响。方法:将63只大鼠随机分为假手术组、模型组和治疗组,每组各21只。采用线栓法构建中动脉栓塞(middle artery embolism,MCAO)模型,假手术组和模型组给予9 ng·L~(-1)氯化钠注射液0.002 mL·g~(-1),加温至37℃直肠灌注。治疗组给予冰黄五苓散0.002 mL·g~(-1),加温至37℃直肠灌注。观察各组大鼠改良神经功能损害评分(modified neurological severity score,mNSS),干湿质量法测定脑组织含水量、脑组织HE染色及免疫组织染色观察神经元特异性烯醇化酶(neuronspecific enolase,NSE)及S-100β的表达情况。结果:模型组mNSS评分高于假手术组,差异具有统计学意义(P <0.05)。治疗组mNSS评分低于模型组,差异具有统计学意义(P <0.05)。假手术组与模型组、治疗组比较,脑水肿含量明显减少,差异具有统计学意义(P <0.05)。治疗组脑组织含水量低于模型组,差异具有统计学意义(P <0.05)。治疗组NSE的D10值明显高于模型组,S-100β的D10值明显低于模型组,差异均有统计学意义(P <0.05)。结论:冰黄五苓散对于大鼠缺血性卒中脑水肿模型具有减缓水肿发展和保护大脑神经功能的作用。
        Objective: To observe the influence of Bing Huang Wuling Powder( BHWLP) for rectal perfusion on neurological function in rats with ischemic stroke.Methods: Sixty-three rats were randomly divided into the sham operation group,the model group and the control group,with 21 rats in each group.Middle artery embolism( MCAO) model was established by thread embolism.The rats in the sham operation group and the model group were given 9 ng·L~(-1)0.002 mL·g~(-1) of sodium chloride injection for rectal perfusion at a temperature of about 37 degrees Celsius.The ones in the treatment group were given 0.002 mL·g~(-1) of BHWLP for rectal perfusion at a temperature of about 37 degrees Celsius.Then each group's modified neurological severity score( mNSS) was observed.The water content in brain tissue was measured by dry-wet mass method.HE staining and immunohistochemical staining of Neuronspecific enolase( NSE) in brain tissue,and S-100 expression were also determined.Results: Compared with the model group and the treatment group,the mNSS score of the sham operation group significantly increased,and the difference was statistically significant( P < 0.05).The mNSS score of the treatment group was lower than that of the model group,and the difference was statistically significant( P < 0.05).Compared with the model group and the treatment group,the content of brain edema decreased significantly in the sham operation group,and the difference was statistically significant( P < 0.05).The water content of brain tissue in the treatment group was lower than that of the model group,and the difference was statistically significant( P < 0.05).The D10 value of NSE in the treatment group was significantly higher than that in the model group,and the D10 value of S-100 beta was significantly lower than that in the model group,and all the differences were statistically significant( P < 0.05).Conclusion: BHWLP has the effect of slowing down edema development and protecting cerebral nerve function in rats with ischemic stroke.
