摘要
目的研究山茱萸中熊果酸的体内、体外抗氧化活性。方法 (1)体外抗氧化实验:用化学反应法,检测熊果酸对邻苯三酚自氧化反应产生的超氧阴离子和Fenton反应产生的羟自由基的清除作用;(2)体内抗氧化实验:昆明种小鼠随机分为空白组、衰老模型组、阳性对照组、低剂量组和高剂量组,每组10只。给药42d后,摘眼球取血,检测小鼠血清中超氧化物歧化酶(SOD)活性和丙二醛(MDA)水平,并测定其胸腺指数和脾脏指数。结果熊果酸具有清除羟基自由基和超氧阴离子自由基的作用,可显著提高小鼠血清中超氧化物歧化酶(SOD)含量、降低丙二醛(MDA)含量,此外,熊果酸组小鼠的胸腺指数和脾脏指数显著升高(P<0.05)。结论熊果酸具有明显的抗氧化活性,为进一步开发中药药材山茱萸提供依据。
        Objective To study the antioxidant activity of ursolic acid from Cornus officinalis in vivo and in vitro.Methods(1) Anti-oxidation experiments In vitro:Application of chemical reaction method to detecting the ursolic acid scavenging effect on superoxide anion from pyrogallol auto-oxidation reaction and the hydroxyl radical of the Fenton reaction.(2) Anti-oxidation experiments In vivo: Kunming mouse were randomly divided into blank control group, aging model group, positive control group, low-dose group and high-dose group, 10 in each group. After the administration of 42 d, pick eyeball blood,detection the mouse of serum superoxide dismutase(SOD) activity and malondialdehyde(MDA) levels, also detection its thymus index and spleen index. Results Ursolic acid has a clear hydroxyl radicals and superoxide anion radical role, it can significantly increase the serum superoxide dismutase(SOD) activity and malondialdehyde(MDA) content, besides, thymus index and spleen index of the Ursolic acid groups mice were significantly increased, P<0.05.Conclusions Ursolic acid has significant antioxidant activity, it provides a basis for the further development of medicinal herbs Cornus.
