摘要
目的:探讨温和灸扳机点干预对局部组织TGF-β1和Smad4表达影响。方法:将54只SD大鼠随机分为A组(空白对照)、B组(模型对照)和C组(模型干预)3组,每组18只;每组又均分3小组(A0-A2、B0-B2和C0-C2)。B、C两组建立肌筋膜痛扳机点模型,并分别给予C1组和C2组温和灸干预。分批处死大鼠,扳机点部取材,制片并进行HE和免疫组化染色,光镜下观察其病理变化。RT-PCR对TGF-β1mRNA及Smad4m-RNA表达水平进行定量分析。结果:TGF-β1、Smad4在正常组织中弱表达,B组和C组强表达;C0组与B0组间TGF-β1mRNA、Smad4mRNA表达量比较无显著差异(P>0.05),C1组与B1组间比较差异有显著意义(P<0.01),C1组较B1组显著下降;C2组与C1组间比较差异有显著意义(P<0.01),C2组较C1组显著下降。结论:温和灸法干预能够下调TGF-β1和Smad4的表达,可能间接调节TGF-β1/Smads信号转导通路的靶基因表达,从而改善扳机点局部肌组织的病理化状态。
        Objective:To investigate the effect of mild moxibustion intervention on local tissue TGF-β1 and Smad4 expression in myofascial trigger points(MTrPs). Method:Fifity-four SD rats were randomly divided into Group A(blank control), Group B(model control) and Group C(model intervention), 18 in each group, and each group was equally divided into 3 groups(A0-A2, B0-B2 and C0-C2). B and C groups set up the MTrPs model, and the C1 group and C2 group were given mild moxibustion intervention. The rats were executed before intervention, intervention for 7 and 15 days. The partial drawing, fixing and making slices. Pathological changes were observed under light microscope after HE and immunohistochmical staining. The expression levels of TGF-β1 mRNA and Smad4 mRNA were quantitatively analyzed by RT-PCR. Results: TGF-β1 and Smad4 were weakly expressed in normal tissues, and strongly expressed in B group and Group C. There was no significant difference in TGF-β1 mRNA and Smad4 m-RNAexpressions between C0 Group and B0 Group(P>0.05). There was significant difference between C1 Group and B1 Group(P<0.01), and C1 group's decreased significantly compared with that of B1 group. There was significant difference between C2 Group and C1 Group(P<0.01), and C2 group's decreased significantly compared with that of C1 group. The C2 group's was significantly lower than that of the C1 group. Conclusion:The mild moxibustion can cut down the expressions of TGF-β1 and Smad4, and the target gene expression of TGF-β1/Smads signal transduction pathway maybe indirectly regulated to improve the pathological state of the local muscle tissue in MTrPs.
